摘 要:目的 :研究黄芪及拆分组分对脾虚水湿不化大鼠的影响,阐明黄芪性味物质基础,为中药性味理论新假说提供依据。方法:采用高脂低蛋白饮食加力竭游泳法建立脾虚水湿不化大鼠模型,以模型大鼠一般状况、血清胃泌素、尿D-木糖、胃排空率、小肠推进率及水负荷指数为观测指标,考察黄芪及拆分组分(黄酮组分、皂苷组分、多糖组分、水提物组分)的作用,进行黄芪性味药理学评价。结果:与模型对照组比较,黄芪水煎液组、各拆分组均可不同程度改善模型大鼠一般状况,升高血清胃泌素含量(P<0.01);黄芪水煎液组、皂苷组和多糖组可提高尿D-木糖排泄率(P<0.05);黄芪水煎液组、黄酮组和多糖组可升高胃排空率(P<0.01),增加小肠推进率(P<0.05,P<0.01);黄芪水煎液组和多糖组可降低水负荷指数(P<0.01)。结论:黄芪能改善脾虚水湿不化模型大鼠脾失健运状况,这可能是其"味甘性温"的功能体现,多糖组分为主要物质基础,其他组分也有部分作用。Objective:To study the effect of astragalus membranaceus and its components on rats with spleen deficiency and water-dampness stagnancy,so as to clarify the property and flavor material basis of astragalus membranaceus and provide basis for the new hypothesis of property and flavor theory of Chinese materia medica. Methods:The rats model of spleen deficiency water-dampness stagnancy was induced by high fat and low protein diet plus swimming exhaustion method. The general condition,serum gastrin levels,urine D-xylose excretion rate,gastric emptying rate,intestinal propulsive rate and water load index were taken as targets for the evaluation of the effect of astragalus membranaceus and its components (flavonoids,saponin, polysaccharide,water extract) and the pharmacologic evaluation of the property and flavor of astragalus membranaceus was made. Result:Compared with model group,water decoction of astragalns membranaceus, flavonoid components,saponin components,polysaccharide components and water extract components all im- proved the general condition and increased the serum gastrin levels of the rats (P〈0.01);water decoction of astragalus membranaceus ,saponin components and polysaccharide components increased the urine D-xy- lose excretion rate(P〈0.05);water decoction of astragalus membranaceus,flavonoid components and polysac- charide components increased the gastric emptying rate(P〈0.01) and the intestinal propulsive rate(P〈0.05, P〈0.01);water decoction of astragalus membranaceus and polysaccharide components decreased the water load index(P〈0.01). Conclusion:Astragalus membranaceus can improve the dysfunction state of spleen in rats with the model. This may be the reflection of its funcion "sweet in taste and warm in nature". The polysaccharide components is the main material basis and other components also have certain effects.
