摘  要:目的 :观察小半夏汤对急性化疗性异食癖大鼠5-HT合成与代谢的影响,探讨小半夏汤防治化疗性恶心呕吐的作用机制。方法:腹腔注射6 mg/kg顺铂建立大鼠化疗性异食癖模型。将Wistar大鼠随机分为正常对照组、小半夏汤正常对照组、顺铂模型组、昂丹司琼阳性药组、小半夏汤大剂量组和小半夏汤小剂量组。各组动物均在造模前1 h和造模后11 h分两次灌胃,昂丹司琼组每日给昂丹司琼2.6 mg/kg,小半夏汤正常对照组和小半夏汤大、小剂量组每日给小半夏汤剂量分别为1.6,3.2,1.6 g/kg,正常对照组和模型组灌胃等容积蒸馏水。每12 h称量一次大鼠摄食高岭土量。造模后24 h处理动物,测定大鼠血清、回肠、延髓5-HT和5-羟吲哚乙酸(5-HIAA)含量以及回肠和延髓色氨酸羟化酶(TPH)、单胺氧化酶A(MAOA)水平。结果:模型组动物摄食高岭土量显著升高,小半夏汤大、小剂量均可抑制化疗大鼠摄食高岭土,均可降低化疗大鼠血清、回肠和延髓5-HT含量以及回肠TPH1和延髓TPH2水平,均可升高化疗大鼠血清、回肠和延髓5-HIAA\5-HT比值以及回肠和延髓MAOA水平。结论 :小半夏汤防治急性化疗性恶心呕吐作用机制与降低回肠与延髓TPH水平、提高MAOA水平,继而抑制5-HT合成、促进5-HT代谢有关。Objective:To observe the effect of Xiao Banxia decoction(XBXD) on 5-HT synthesis and metabolism of rats with acute chemotherapy-induced pica and explore the mechanism of XBXD on prevention and treat- ment of chemotherapy-induced nausea and vomiting. Methods:The chemotherapy-induced pica rat model was established by intraperitoneal injection of cisplatin(6 mg/kg). The Wistar rats were randomly divided into normal control group,XBXD normal control group,cisplatin model group,positive group of ondansetron,high dose of XBXD group and low dose of XBXD group. All rats were given gavage respectively at 1 hour be- fore and 11 hours after the model establishment. The rats in ondansetron group were given ondansetron twice a day,2.6 mg/kg/d. The rats in XBXD normal control group,XBXD high and low dose groups were given XBXD,respectively 1.6,3.2 and 1.6 g/kg/d. The rats in normal control group and model group were given equal volume of distilled water by gavage. Kaolin consumptions of rats were weighed every 12 hours. 24 hours after model establishment,the contents of 5-HT and 5-HIAA in rats' serum,ileum and medulla oblongata and levels of TPH and MAOA in ileum and medulla oblongata were measured. Results:The Kaolin consumptions of rats in model group increased significantly. Both high and low dose of XBXD could significantly inhibit the kaolin consumptions,reduce 5-HT contents in serum,ileum and medulla oblon- gata,TPH1 level in ileum,TPH2 level in medulla oblongata and increase the ratios of 5-HIAA/5-HT in serum,ileum and medulla oblongata and MAOA level in ileum and medulla oblongata. Conclusion: The mechanism of XBXD on the prevention and treatment of acute chemotherapy-induced nausea and vomiting was related to the decrease of TPH level and the increase of MAOA level in ileum and medulla oblongata which can suppress the synthesis of 5-HT and promote its metabolism.
