摘  要:采用M-csF+RANKL联合诱导RAW264.7细胞分化为破骨样细胞(OCL),观察培养过程中的凋亡现象,了解其生长规律。在诱导培养的第5~9天,通过TRAP染色、检测TRAP基因表达水平、细胞骨架与胞核染色方法分析OCL凋亡状况。结果表明:体外培养成熟OCL短时间内即发生凋亡,表现为TRAP阳性细胞数减少,TRAP基因表达水平降低,细胞骨架结构破坏、胞核形态改变。Osteoclast-like-cells (OCLs) were induced from RAW264.7 via M-CSF+ RANKL, the apoptosis of OCLs were investigated, and the growth rhythm of OCLs were observed. TRAP staining, TRAP gene expression level assay, cytoskeleton and nuclei staining were employed to study the apoptosis of OCL from 5th to 9th d. The results indicated that mature OCL apoptosis developed in a short term in vitro. The apoptotic OCL was characterized by decline in TRAP- positive cells number, reduction in TRAP gene expression level, destruction of cytoskeleton and morphological changes of nuclei.
