摘 要:为了建立可以快速对海狸鼠肉进行鉴别的特异性PCR方法,试验采用线粒体Cyt B基因序列设计引物,对海狸鼠肉进行特异性扩增,并对此PCR方法进行条件优化。结果表明:海狸鼠可扩增出305 bp的条带,且对其他常见物种无反应;引物浓度、退火温度和Mg2+浓度对反应结果影响不大;当模板稀释100倍时仍有清晰条带;对肉品进行高温高压处理以及掺杂处理均不影响扩增结果。说明此方法可以应用于海狸鼠的物种鉴别。To establish a specific PCR assay for the rapid identification of nutria meat, the primers were designed using the sequence of mito- chondrial cytochrome b gene. The specific amplification was carried out in the nutria meat, while the conditions of PCR assay were optimized. The results showed that a 305 bp band was amplificated from the nutria meat, and there were no reactions to other common species. The primer concentrations, annealing temperature and the concentration of Mg2+ had little effect on the reaction result. There was still a clear band when the DNA template was diluted 100 times. The meat treated with high temperature and pressure and the mixed treatment, had no effects on the amplification result. The results indicate that this assay can be used for the species identification of nutria meat.
