摘 要:以胸腺五肽(Tp-5)为模板分子、丙烯酰胺(AM)为功能单体、乙二醇二甲基丙烯酸酯(EGDMA)为交联剂、羧基功能化Fe3O4磁性粒子为载体,采用乳液聚合法制备出胸腺五肽分子印迹聚合物磁性微球(Tp-5-MIPMMs)。通过研究交联剂用量、预聚合时间、聚合反应时间对Tp-5-MIPMMs吸附性能和印迹因子的影响,确定了最佳制备条件。扫描电镜(SEM)分析显示微球表面被多孔、均匀聚合物包覆,粒径40μm左右。静态和动态吸附研究表明Tp-5-MIPMMs在2.0 g·L-1的Tp-5溶液中吸附8 h达到饱和,最大吸附量达15.63 mg·g-1,印迹因子达1.51。以谷胱甘肽(GSH)作对比,用薄层色谱(TLC)研究了Tp-5-MIPMMs的选择吸附性能,结果表明Tp-5-MIPMMs对Tp-5分子具有特异选择能力。Thymopentin molecularly imprinted polymer-coated magnetic microspheres (Tp-5-MIPMMs) with selective adsorption of thymopentin were prepared by emulsion polymerization on the surface of carboxyl-functionalized Fe3O4 using thymopentin as a template molecule, acrylamide as a functional monomer and ethylene glycol dimethacrylate as a cross-linker. The optimum conditions for preparing Tp-5-MIPMMs were obtained by investigating the effects of cross-linker percentage, pre-polymerization time and polymerization time on adsorption performance and imprinting factors. Scanning electron microscopy (SEM) results indicate that the magnetic microspheres with diameter of- 40 μm are uniformly coated by porous MIPs layers. Static and dynamic adsorption results show that the adsorption equilibrium is 15.63 mg.g^-1 with an imprinting factor of 1.51. The specific selectivity of Tp-5-MIPMMs was evaluated by thin layer chromatography (TLC) with glutathione (GSH) as analogs and the result shows the Tp-5-MIPMMs has excellent thymopentin selectivity.
