断奶后仔猪肠壁Obestatin的定位及定量研究

摘　　要：为了测定肥胖抑制素(Obestatin)在断奶后仔猪肠壁中的分布状况以及表达量,对断奶15 d仔猪(45日龄)处死后立即取十二指肠、空肠、回肠、盲肠、结肠和直肠。一部分组织用于固定并制作组织切片,利用免疫组化试剂盒对切片进行检测,并拍照分析;另一部分组织用于提取总蛋白并用ELISA试剂盒检测各肠段组织Obestatirt蛋白浓度。结果断奶后仔猪肠道均有Obestatin的广泛分布,且不同组织其表达量不同。空肠Obestatin表达量最低,显著低于大肠(P<0.05);大肠(结肠与直肠)表达量高于其他肠道组织。表明Obestatin在仔猪肠壁中确实存在,且在肠壁不同部位的表达量有所不同。The experiment was conducted to study the distribution and contents of Obestatin in the intestinal wall of weanedpiglets by the methods of immunohistochemisty and ELISA.The resuhs showed that Obestatin was widely and differently expressedin different intestinal tissues of weaned piglets.The concentration of Obestatin protein was at the lowest level in jejunum and its lev-el was significantly lower than that in large intestine （colon and rectum）（P〈0.05 ）. In conclusion, Obestatin did exist in intestinalwall of weaned piglets, and its levels of expression were different in different parts of the intestine.
