摘  要:翻译调控肿瘤蛋白(TCTP)是一类广泛存在于真核生物中且具有多种重要生物学功能的蛋白质家族。利用RT-PCR和RACE技术从广东桑(Morus atropurpurea)品种中桑5801号植株的根部克隆了一种TCTP蛋白的编码基因,命名为Ma-TCTP(Gen Bank登录号:KP228013)。Ma-TCTP基因c DNA全长841 bp,包括507 bp的ORF以及101 bp的5'-UTR和233 bp的3'-UTR,ORF编码168个氨基酸,蛋白质分子质量为18.736 2 k D,等电点为4.53。实时荧光定量PCR检测Ma-TCTP基因在桑树的根部、茎部和叶片中均有表达,其中在根部组织中的表达量最高,茎中次之,叶片中最低。在低温(4℃)和高温(35℃)胁迫下,Ma-TCTP基因在桑树根部、茎部和叶片组织的表达量均大幅上升,其中在叶片中的表达量升高最为明显,推测Ma-TCTP基因可能参与桑树对温度逆境胁迫的生理应激响应过程。The translationally controlled tumor protein (TCTP) is widely expressed in all eukaryotic organisms and has a variety of biological functions. In this study, a TCTP protein coding gene was cloned from the roots of Zhongsang 5801 of Morus atropurpurea by RT-PCR and RACE methods and was named as Ma-TCTP (GenBank accession No. KP228013). Full length cDNA of Ma-TCTP gene was 841 bp including 507 bp open reading frame (ORF), 101 bp 5'-UTR and 233 bp 3'-UTR. The ORF encoded a protein of 168 amino acids with a calculated molecular weight of 18. 736 2 KD and isoelectric point of 4. 53. Real-time fluorescent quantitative PCR revealed that Ma-TCTP gene was expressed in root, stem, and leaf. The expression level of Ma-TCTPwas the highest in root followed by stem, and was the lowest in leaf. The expression level of Ma-TCTP in root, stem and leaf increased greatly under stress of low temperature (4 ℃) and high temperature (35 ℃). Especially, the increase of expression level in leaf was the most obvious, suggesting that Ma-TCTPgene may participate in the temperature stress response of mulberry.
