家蚕烟酰胺腺嘌呤二核苷酸激酶基因(BmNADK)的高温应激表达分析

摘  要:烟酰胺腺嘌呤二核苷酸激酶(NAD kinase,NADK)是生物体内氧化还原调节和氧化应激反应的重要酶类。以对高温耐受性不同的家蚕品种的5龄幼虫为材料,进行34℃连续高温处理和42℃高温冲击3 h处理,通过实时荧光定量PCR检测家蚕NADK基因(Bm NADK)在雌蚕和雄蚕的脂肪体、中肠、丝腺3个器官组织中的表达水平,探究该基因是否与家蚕对高温的应激反应有关。结果显示,5龄期2种高温环境胁迫均可引起Bm NADK基因在蚕体3个器官组织中上调表达,其中在脂肪体表达的上调最为显著,且雄蚕脂肪体中的上调表达明显高于雌蚕,当恢复常温环境条件时,Bm NADK基因在蚕体组织的表达量也随之下降;高温胁迫下Bm NADK基因在耐高温家蚕品种7532幼虫组织中的表达量高于对高温耐受性差的品种皓月。研究结果显示Bm NADK基因与家蚕对高温的应激反应相关,在耐高温家蚕品种及雄蚕体内的应激表达水平相对较高。Nicotinamide adenine dinucleotide kinase (NAD kinase, NADK) is an important enzyme that involves the red- ox regulation and oxidative stress. In order to investigate whether silkworm ( Bombyx mori) NADK gene (BmNADK) is re- lated to stress response, the 5th instar larvae of silkworm varieties with different high temperature tolerance were used as materials. Under continuous high temperature treatment at 34 ℃ and after heat shock at 42 ℃ for 3 h, the expression lev- els of BmNADK in fat body, midgut and silkgland of female and male silkworm larvae were detected by real time fluores- cent quantitative PCR (qRT-PCR). The results showed that the expression levels of BmNADK gene in all three tissues of the 5th instar was up-regulated under two kinds of high temperature stress, and the most distinctive increase was observed in fat body. Meanwhile, the expression level of BmNADK gene in fat body of male larvae was higher than that in female larvae. When temperature decreased to normal level, the expression levels of BmNADK gene in three tissues dropped as well. Under high temperature stress, the expression levels of BmNADK gene in tissues of high temperature tolerance silkworm variety 7532 were higher than those of high temperature sensitive variety Haoyue. The above results indicated that BmNADK gene in silkworm is related with its tolerance to heat stress, and the expression levels of BmNADK is relatively higher in high temperature tolerance silkworm variety and male larvae.
