摘 要:目的探讨补肾温阳中药通过诱导转录因子Foxp3活化、调控调节性T细胞,而发挥对变应性哮喘的免疫治疗作用。方法80只BALB/c(白变种实验室老鼠)小鼠随机分成4组:对照组、哮喘组、中药干预组及中药+抗体干预组,用屋尘螨(HDM)变应原经腹腔注射致敏、滴鼻激发复制变应性哮喘样肺部炎症模型。中药干预组、中药+抗体干预组每只小鼠给予补肾温阳中药(用淫羊藿、巴戟天免煎颗粒配制500 ul灌胃qod,对照组给予500 ul NS。观察各组支气管肺泡灌洗液(BALF)中细胞分类及计数结果;检测各组BALF上清细胞因子IL-10、IFN-γ、TGF-β1浓度及血清Ig E水平;分离小鼠脾脏细胞,以流式细胞仪方法检测CD4+CD25+Treg细胞;用实时荧光定量PCR检测Foxp3-m RNA。结果正常对照组、中药干预组BALF中IL-10、IFN-γ、TGF-β1浓度分别高于哮喘模型组(P<0.01)和中药+抗体干预组(P<0.05);中药干预组BALF中IL-10、IFN-γ、TGF-β1浓度低于正常对照组(P<0.05);哮喘模型组BALF上清细胞因子(IL-10、IFN-γ、TGF-β1)浓度高于中药+抗体干预组(P<0.05)。血清Ig E水平正常对照组、中药干预组均分别低于哮喘模型组(均为P<0.01)及中药+抗体干预组(均为P<0.05);中药干预组血清Ig E水平低于正常对照组(P<0.05)。哮喘模型组小鼠脾脏中CD4+CD25+调节性T细胞数量、Foxp3-m RNA表达水平较正常对照组、中药干预组均明显减低,差异有统计学意义(分别为P<0.01,P<0.05)。与哮喘模型组、中药+抗体干预组比较,中药干预组小鼠脾脏中CD4+CD25+调节性T细胞数量、Foxp3-m RNA表达水平显著增高,差异有统计学意义(均为P<0.05)。与正常对照组比较,中药干预组小鼠脾脏中CD4+CD25+调节性T细胞数量、Foxp3-m RNA表达水平较低,差异有统计学意义(P<0.01)。与哮喘模型组比较,中药+抗体干预组小鼠脾脏中CD4+CD25+调节性T细胞数量、Foxp3-m RNA表达水平降低,差异有统计学意义(均为P<0.05)Objective o investigate the effects of the traditional Chinese medicine which is tonifying kidney and warming Yang on allergic asthma immunotherapy by inducing the activation of the transcription factor Foxp3 and regulating the Treg. Methods The 80 BALB/c mice were randomly divided into four groups: the control group, the Asthma group, the traditional Chinese medicine intervention group and the traditional Chinese medicine and antibodies intervention group. Asthma model was established by injecting HDM allergen via in- traperitoneal injection and nasal inhalation to evoke inflammation in lung. The boil-free granule of Epimedium Herb and Morinda offici- nalis, which has the effect of tonifying kidney and warming yang, is prepared into herbal extracts. An amount of 500 ul of herbal extract- sare used in the traditional Chinese medicine intervention group and the traditional Chinese medicine and antibodies intervention group by intragastric administration, while 500ul NS are used in the control group. The cell types and amounts in each group's BALF were ob- served. The concentrations of cytokine IL-IO, IFN-'y and TGF-β1 in BALF' supernate and the serum IgE level were tested. Meanwhile, detect CD4CD25regs in spleen cells of mieeby by flow cytometry and the level of Foxp3-mRNA by real-time PCR. Results The con- centrations of IL-10, IFN-y, TGF-β1 in BALF of the control group and the traditional Chinese medicine intervention group were respec- tively higher than those of the asthma model group (P〈0.01) and the traditional Chinese medicine and antibodies intervention group (P〈 0.05) . The concentrations of IL-10, IFN-y and TGFq31in the traditional Chinese medicine intervention group were notbly lower than those of the control group (P〈0.05) . The concentrations of supernatant cytokine (IL-10, IFN-y, TGF-β1) in BALF of asthma model group were significantly higher than those of the Chinese herbcide and antibodies intervention group (P〈0.05) .
