摘  要:目的研究六君子汤对食管癌血管生成的影响。方法 MTT法检测六君子汤醇提物对食管癌细胞株EC9706生长抑制作用,收集其500μg/m L质量浓度的EC9706细胞条件培养基用于鸡胚绒毛尿囊膜的血管生成、人脐静脉内皮细胞增殖、迁移和小管形成的观察。ELISA法检测EC9706细胞和人脐静脉内皮细胞培养基上清中VEGF和IL-6含有量。结果六君子汤醇提物可明显抑制EC9706细胞增殖,具一定剂量依赖性,其IC50值为505.28μg/m L。六君子汤醇提物可减少EC9706细胞条件培养基所致鸡胚绒毛尿囊膜血管生成、人脐静脉内皮细胞增殖、迁移和小管形成。六君子汤醇提物可减少EC9706细胞和内皮细胞IL-6及内皮细胞VEGF分泌。结论六君子汤醇提物可能通过干预EC9706细胞信号通路从而抑制血管生成的效应。AIM To study the effects of Liujunzi Decoction on the angiogenesis of esophageal cancer cell line EC9706. METHODS The inhibitory effect of Liujunzi Decoction alcohol extract on proliferation of EC9706 was detected by MTY assay. 500μg/mL of EC9706 cell conditioned media in Liujunzi Decoction was collected and used to observe the angiogenesis of chick embryo chorioallantoic membrane (CAM), proliferation migration and tube formation of human umbilical vein endothelial cells (HUVECs). VEGF and IL-6 level in culture medium su- pernatant of EC9706 cells and HUVECS were detected by ELISA. RESULTS Liujunzi Decoction alcohol extract could inhibit the proliferation of EC9706 cells with IC50 value of 505.28μg/mL in a dose dependent manner, and markedly reduce the elevation in the angiogenesis of CAM, HUVECs proliferation, migration and tube formation caused by EC9706 cell conditioned medium. It also decreased both IL-6 level from EC9706 and HUVECs and VEGF level from HUVECs in serum. CONCLUSION Liujunzi Decoction alcohol extract can inhibit the angio- genesis through intervening secretion of EC9706 cell signal pathway.
