三种口腔溃疡中药制剂对小鼠成纤维细胞生长增殖和胶原合成的影响

摘　　要：目的探讨3种治疗复发性口腔溃疡中药制剂(西瓜霜、云南白药、甘草泻心汤)在细胞水平上促进溃疡愈合的作用机制。方法建立小鼠口腔成纤维细胞体外培养模型,以华素片为对照,采用MTT、Elisa测定3种中药制剂对成纤维细胞增殖、胶原合成的影响。结果西瓜霜在促进成纤维细胞的增殖及胶原合成,抑制MMP-2,促进TIMP-1方面有较显著的作用。云南白药促进能够HYP的分泌和抑制MMP-2的分泌。甘草泻心汤对成纤维细胞有一定的抑制作用。结论提示临床上治疗复发性口腔溃疡时应联合用药或分期用药。AIM To study the mechanisms of three kinds of herbal preparations （ Watermelon Frost, Yunnan Baiyao, Gancao Xiexin Decoction） for recurrent oral ulcer on ulcer healing in cellular levels. METHODS Mice fibroblast L929 was used as the in vitro model for recurrent oral ulcer; cydiodin was employed as the control drug. The effects of herbal preparations on the fibroblast proliferation and collagen production were determined by MTT assay and Elisa Kit, respectively. RESULTS Watermelon Frost promoted the fibroblast proliferation and collagen synthesis, inhibited MMP-2 level and increased TIMP-1 level; Yunnan Baiyao stimulated the secretion of HYP and down-regulated the secretion of MMP-2 level ; Gancao Xiexin Decoction inhibited the fibroblast. CONCLUSION The experimental data suggests that the clinical treatment for recurrent oral ulcer should be approached by combi- ning these three preparations or applying them in different stages.
