摘 要:目的 考察合煎、分煎抗病毒方银黄制剂提取液中总黄酮、绿原酸、黄芩苷的变化,从而为其临床应用提供依据.方法 采用紫外-可见分光光度法测定总黄酮;采用反相液相色谱法,Diamonsil (TM)钻石C18柱,5μm(250 mm×4.6 mm);流动相分别以乙腈-0.4%-磷酸(13:87),在327 nm波长处测定绿原酸;甲醇-水-磷酸(47∶ 53∶0.2)在280 nm波长处测定黄芩苷.结果 总黄酮在0.112~0.56 mg、绿原酸在1.14~5.70 μg,黄芩苷0.44~2.20μg范围内线性关系良好,分煎液中总黄酮和黄芩苷的含量明显高于合煎液,合煎液和分煎液中绿原酸的量无明显差别.结论 银黄复方制剂合煎和分煎对有效成分的溶出有影响.Objective To compare the contents of total flavonoids,chlorogenic acid,baicalin in single medicine decoction and mixed medicine decoction of Yinhuang preparations,so as to provide a scientific basis for clinical application of Yinhuang preparations.Methods Ultraviolet spectrophotometry was used to determinate total flavonoids,chlorogenic acid and baicalin were determined by HPLC.Diamonsil-C 18 column was used,chlorogenic acid was detected at 327 nm and mobile phase was consisted of acetonitrile-0.4% phosphoric (13∶87).Baicalin was detected at 280 nm,and mobile phase was methanolwater-phosphoric acid (47∶53∶0.2).Results The total flavonoids,chlorogenic acid and baicalin all had good linear relationship.The contents of total flavonoids and baicalin in single medicine decoction were obviously higher than those in mixed medicine decoction,and the content of chlorogenic acid had no obvious change.Conclusion The extraction of active components of Yinhuang preparations of different decoctions changes a lot.
