摘 要:目的探索从人类口腔黏膜上皮细胞中提取基因组DNA方法的可行性。方法分别采用煮沸法和碱裂解法从人口腔黏膜上皮细胞提取基因组DNA,然后与临床常用的碘化钠(NaI)法提取的人全血基因组DNA进行比较。结果煮沸法和碱裂解法2种方法均能从口腔黏膜上皮细胞抽提到DNA,提取的DNA浓度虽低于NaI法,但用2种方法提取到的DNA样品进行聚合酶链反应,均能获得满意效果,与常规NaI法无明显差异。结论从人口腔黏膜上皮细胞提取基因组DNA简便、快捷、无损,在临床上具有潜在的应用价值。Objective To explore the feasibility of methods of extracting genomic DNA from human oral epithelial cells .Methods The alkaline lysis and boiling were used to extract the genomic DNA from human oral mucosal epithelial cells .Then the methods were compared with the traditional method of sodium iodide (NaI) ,in which DNA was extracted from human whole blood . Results The genomic DNA could be extracted from oral mucosal epithelial cells by both methods ,which had a lower concentration . But the polymerase chain reaction amplification were successful and there was no significant differences among all methods .Conclu‐sion The methods of extracting genomic DNA from the human oral mucosal epithelial cells is simple ,efficient and harmless ,which has potential value of clinical applications .
