摘 要:目的研究马钱子碱透皮贴剂对小鼠和大鼠的镇痛作用及对大鼠脑啡肽含量的影响。方法 (1)小鼠的镇痛实验采用电刺激法,将50只小鼠随机分成空白对照组、芬太尼组及马钱子碱透皮贴剂小、中、大剂量组,分别给药后,用电刺激法观察给药后20 min及1、2、4、8、10 h不同时间点小鼠痛阈值的变化;(2)大鼠的镇痛实验采用5-HT致痛法,将50只大鼠随机分成5组,分组方法同前,分别给药后30 min给予5-HT致炎,用足压痛法观察给药后1、2、4、8 h不同时间点大鼠致炎足的痛阈值的变化。(3)将另50只大鼠随机分成5组,分组方法同前,给药后30 min分离大鼠脑功能区,采用放射免疫分析法观察给药后大鼠脑内脑啡肽的含量的变化。结果马钱子碱贴剂能提高小鼠电刺激致痛的痛阈值,能提高5-HT致痛的大鼠镇痛率和大鼠大脑功能区脑啡肽的含量。结论马钱子碱透皮贴剂对小鼠电刺激和大鼠5-HT致炎引起的疼痛有一定的镇痛作用,其作用的机制与增加脑功能区脑啡肽的含量有关。Objective To investigate the effects of brucine transdermal patches on analgesia and enkephalins content in rats. Methods (1) The analgesic experiments of mice were adopted by electric stimulation method. 50 mice were randomly divided into the blank control group, fentanyl group, and brucine transdermal patches groups (high, medium and low doses). The pain thresholds were observed at 20 min, 1, 2, 4, 8 h after medication by electrical stimulation. (2) The analgesia experiment of rats was tested by 5-HT. The 50 rats were randomly divided into 5 groups, and the grouping method was same with before. The rats were caused inflammatory by 5-HT after 30 minutes of given medicine. The pain thresholds of rats at 1, 2, 4, 8 h were observed by foot tenderness. (3) Another 50 rats were randomly divided into 5 groups, the brain functional region were separated after 30 minutes of medication. The changes of enkephalins content in rats were observed by radioimmunoassay. Results Brucine transdermal patches could increase the pain thresholds by electrical stimulation, analgesic rates of rats by 5-HT and enkephalins content in rats. Conclusion Brucine transdermal patches showed the analgesic effect on the pain caused by electrical stimulation and 5-HT and its mechanism was related with the contents of enkephalin in brain functional area.
