摘 要:目的观察电针"大椎穴"对颈椎病模型大鼠椎间盘纤维环细胞Wnt1蛋白、糖原合成酶激酶-3β(glycogen synthase kinase-3β,GSK-3β)的影响,并初步探讨电针对颈椎间盘退变调控及抑制纤维环细胞凋亡的机制。方法选用SPF级SD大鼠40只,随机分成空白组、模型组、电针组、美洛昔康组,采用Western blot法检测颈椎间盘的纤维环细胞Wnt1、GSK-3β蛋白的表达水平的改变。结果模型组与空白组比较差异有统计学意义(P<0.05);电针组、美洛昔康组与模型组比较差异有统计学意义(P<0.05)。电针可增加颈椎间盘纤维环细胞的阳性表达,显著上调Wnt1、GSK-3β蛋白表达的水平。结论电针"大椎穴"可能是通过调节经典的Wnt/β-catenin信号通路中关键因子Wnt1蛋白、GSK-3β蛋白的表达来抑制颈椎病模型大鼠的椎间盘纤维环细胞凋亡。Objective To observe the effect of electroacupuncture(EA) at Dazhui point on Wnt1 and glycogen synthase kinase-3β(GSK-3β) in vertebral disc cells of cervical spondylosis model rats and explore its mechanism in relieving intervertebral disc degeneration and inhibition of fiber ring cell apoptosis. Methods 40 SPF SD rats were randomly divided into the blank group, model group,EA group and meloxicam tablets group(MT group). The expression of Wnt1 and GSK-3βwere detected by Western blot. Results Comparing the model group with blank group was statistically significant(P 0.05).Compared with the model group, differences of the EA and MT groups are statistically significant(P〈0.05). EA could increase the positive expression of vertebral disc cells, and up-regulate the level of Wnt1 and GSK-3β. Conclusion EA of Dazhui point can effectively relieve the degeneration degree of cervical spondylosis model rats, which is probably related to its effect in regulating the expression of key factors Wnt1 and GSK-3β in the classical Wnt/β-catenin signal pathway.
