摘 要:以东北红豆杉(Taxus cuspidata Sieb.et Zucc.)果实成熟后的种子为供试材料,通过多因子交叉重复试验对影响东北红豆杉种胚发育的主要因素及其水平进行了探讨,旨在筛选并确定调控东北红豆杉种胚形态发育的主要因素及其适宜水平。结果表明,东北红豆杉种胚发育的调控方法和主要因素及其水平为以饱和碳酸钠溶液浸泡种子20-30 min除油脂后用粗沙去除部分种皮,再用1.80 mg/L 6-苄氨基腺嘌呤(6-BA)、2.20 mg/L赤霉素(GA3)和0.30 mg/L 2,4-二氯苯氧乙酸(2,4-D)混合溶液浸泡36 h,将种子与河沙(1∶3)混合,进行155 d的变温处理,变温幅度和时间:白天温度为(23±2)℃,处理10 h,夜间温度为(15±2)℃,处理14 h,子叶胚完全分化率98.0%,种子发芽率95.0%以上。The seeds in fruit ripening of Taxus cuspidata were used to study the main factors with different levels for development of seed embryonic of T. cuspidata through repeated experiments with multi factorial and cross, in order to screen and establish the main factors and suitable levels to control morphological development of seed embryonic of T. cuspidata. Results showed that the method and the main factors with levels for development of seed embryonic of T. cuspidata were to soak the seeds with saturated solution of sodium carbonate in 20-30 min, grinding the part of seed coat with coarse river sand, and using the mixed solutions with 1.80 mg/L 6-BA, 2.20 mg/L GA3 or 0.30 mg/L 2,4-D in 36 h. The seeds and sand(1:3)were mixed, treated with temperature change in 155 d. The extent of temperature change and time were (23±2)℃ and 10 h in day, (15±2)℃ and 14 h in night. The fully differentiated rate of cotyledonary embryo was 98.0%. The germination rate was 95.0%.
