摘  要:正常细胞型朊蛋白(PrPC)是一种高度保守、在所有哺乳动物体内广泛表达的细胞表面糖蛋白,人类PrPC在胚胎发育早期即已表达。研究表明,PrP在多种癌症中表达,包括胃癌、胰腺癌、大肠癌、乳腺癌、前列腺癌、肝癌(HCC)、口腔鳞状细胞癌(OSCC)等,影响这些癌症的发生与侵袭,并在多药耐性(MDR)获得中起重要作用。因此,朊蛋白有望成为治疗多种癌症的新靶标。论文就PrP的结构、功能及其在细胞功能和疾病发生发展中的作用,以及PrP在多种肿瘤发生发展、耐药性的作用等研究进行概述,并对PrP在癌症新型疗法未来发展中的潜在影响进行分析讨论,以期为PrP相关肿瘤病的防治方面做出贡献。To explore the significance of prion protein(PrP) in the occurrence and development of cancer,the current research progresses on the structure, function of PrP and it's role in cell function, disease and in the development, progression, and drug resistance of various cancers were viewed, and it's potential effects for future development of new therapeutic treatments of cancer were discussed, to contribute to the pre- vention and treatment of PrP related cancers. The normal cellular prion protein(PrPc) is a highly conserved and widely expressed cell surface glycoprotein in all mammals, which is generally located on the cell mem- brane associated with cholesterol-rich microdomains (rafts) in cultured non-neuronal and neuronal cells. Human PrPc is expressed early in embryogenesis. In adults, the highest level of PrPc expression is ob- served in neurons of the brain and spinal cord, specifically in those of the cerebellum, cerebral cortex, thalamus, hippocampus and medulla oblongata. Numerous research data show that PrP overexpresses in various cancers, including gastric cancer, pancreatic cancer, colorectal cancer, breast cancer, prostate cancer, hepatocellular carcinoma(HCC) and oral squamous cell carcinomas(OSCC) influence the occur- rence and incursion of these cancers, and play important roles in acquisition of drug tolerance (MDR) of gastric cancer. Therefore, PrP is expected to become a new therapeutic target, and to open new doors for the treatment of various cancers.
