摘 要:目的:研究湿热因素对结肠癌生长、血管新生和转移的影响。方法:在建立小鼠结肠癌原位模型的基础上,采用复合因素的方法建立小鼠湿热结肠癌模型。第41天处死小鼠,测量瘤体质量;每组剩余6只小鼠用于观察生存时间;免疫组化法检测Ki67和微血管密度(MVD)表达;ELISA测定血清中血管内皮细胞生长因子(VEGF)、基质金属蛋白酶-2(MMP-2)和基质金属蛋白酶-9(MMP-9)表达。结果:本研究以中医理论为指导成功建立了湿热结肠癌小鼠模型。湿热对结肠癌肿瘤的生长没有明显影响,但能够促进结肠癌的肝转移率,湿热证结肠癌小鼠组的生存时间明显缩短。免疫组化结果显示,湿热对结肠癌组织Ki67表达没有明显影响,但能够促进肿瘤MVD形成。ELISA结果显示,湿热能够上调荷瘤小鼠血清中VEGF、MMP-2和MMP-9表达水平。结论:湿热对结肠癌的生长没有明显影响,但能够促进结肠癌的肝转移,其机制可能与湿热上调VEGF、MMP-2和MMP-9表达有关。Objective: To investigate the mechanism of the promoting effect of dampness-heat on tumor growth, angiogeuesis, and metastasis of colonic cancer. Methods: The dampness-beat colonic cancer mouse model was established using an orthotopic colonicl cancer model. Part of the mice were sacrificed at day 41 and tumor weights were measured; the remaining mice (6 mice in each group) were maintained longer for survival studies. The expressions of Ki67 and MVD were tested by immunohistochemistry. ELISA was utilized to quantify the serum levels of VEGF, MMP-2 and MMP-9. Results: In this study, we successfully established a traditional Chinese medicine dampness-heat colonic cancer mouse model. Dampness-heat had no impact on the colonic cancer tumor growth. However, dampness-heat significantly enhanced the liver metastasis of colonic cancer, resulting in shorter survival time for the dampness-heat mice. Compared with the colonic cancer group, Ki67 expressed was in similar levels in dampness-heat colonic cancer group. The MVD in the dampness-heat group was significantly higher than the colonic cancer group. Moreover, dampness-heat could increase serum levels of VEGF, MMP-2 and MMP-9. Conclusion: In spite of no impact on tumor growth, dampness-heat significantly heightens colonic cancer metastasis to the liver, via a probable mechanism of enhancing VEGF, MMP-2 and MMP-9 expression.
