摘 要:目的:观察黄芪多糖(APS)对四氯化碳(CCl4)诱导肝纤维化大鼠转化生长因子-β1(TGF-β1)/Smads信号通路的影响。方法:采用CCl4诱导大鼠肝纤维化模型,取大鼠血清检测肝功能;Masson染色比较肝纤维化程度;免疫组化法测肝组织TGF-β1蛋白表达;Real-time PCR分析肝组织TGF-β1、Smad3、Smad4以及Smad7的表达。结果:与模型组比较,APS各剂量组的血清肝功水平、肝纤维化程度明显降低,肝组织TGF-β1、Smad3、Smad4表达量显著降低,Smad7表达量显著增高。结论:APS对CCl4诱导的大鼠肝纤维化有显著的干预作用,其作用机制可能与调节TGF-β1/Smads信号通路有关。Objective: To investigate effects of astragalan on liver fibrosis rat induced by carbon tetrachloride (CCl4) in TGF-β1/Smads signal pathway. Methods: The rat model of experimental liver fibrosis was established by CCI4, and liver function was tested by serum of rat. The comparition of the fibrosis degrees was observed using Masson staining. The protein expressions of TGF-β1/Smads was detected by immunohistochemistry method; The mRNA of TGF-β1, Smad3, Smad4 and Smad7 were assessed using Real-time PCR. Results: Compared with the model group, the level of liver function and the degree of fibrosis in serum of Alas group lowered obviously, the expression of TGF-β1, Smad3 and Smad4 was significantly decreased, and the expression of Smad7 was significantly increased. Conclusion: Astragalan has significantly interfering effect on liver fibrosis rat induced by carbon tetrachloride (CCl4), and its mechanism may be related to regulation TGFq3/Smad signaling pathway.
