摘 要:目的:观察四物汤对血虚证小鼠肝细胞凋亡及凋亡相关基因Bax、Bcl-2和Fas表达的影响。方法:30只C57BL/6J小鼠随机分为正常组、模型组和给药组,每组10只。给药组予四物汤灌胃20g·kg-1·d-1,正常组和模型组则予以等体积0.9%氯化钠溶液。以腹腔注射环磷酰胺建立血虚证模型,采用全自动血液分析仪检测外周血象,TUNNEL法检测肝细胞凋亡,荧光定量PCR法检测Bax、Bcl-2及Fas m RNA表达。结果:与模型组相比,给药组小鼠外周血红细胞数、白细胞数以及血红蛋白含量升高(P<0.05),血小板数无统计学差异,肝细胞凋亡百分率降低(P<0.05),促凋亡基因Bax、Fas m RNA表达水平下降(P<0.05),抗凋亡基因Bcl-2 m RNA表达无统计学差异。结论:四物汤可改善血虚证模型小鼠外周血象,下调促凋亡基因Bax和Fas表达,减少肝细胞凋亡。提示四物汤补益肝血作用机制可能与其减少肝细胞凋亡有关。Objective: To observe the influence of Siwu Decoction on hepatocyte apoptosis and related genes expression such as Bax, Bcl-2 and Fas in mice with blood deficiency. Methods: 30 C57BL/6J mice were randomly divided into control group, model group and treatment group. Rats in treatment group received gavage administration with Siwu Decoction, and rats in control and model group received gavage administration with normal saline in the same volume as Siwu Decoction. Blood deficiency mouse models were established by intraperitoneal injection of cyclophosphamide. Automated hematology analyzer was used to test the periphery blood parameters, and Tunnel method was used to test the hepatocyte apoptosis. The mRNA expression of Bax, Bcl-2 and Fas was tested by using RT-PCR. Results: Compared with model group, the number of red blood cell and while blood cell and hemoglobin in peripheral blood of mice in treatment group were increased significantly (P〈0.05), and hepatocyte apoptosis rate and the mRNA expression level of Bax, and Fas were decreased (P〈0.05), while the difference on platelet count and mRNA expression level of Bcl-2 were not significant. Conclusion: Siwu Decoction could improve the periphery blood hemogram, decrease the expression of Bax and Fas, and induce hepatocyte apoptosis. It showed that the mechanism of replenishing and nourishing liver blood of Siwu Decoction might be related to reducing hepatocyte apoptosis.
