摘 要:目的观察POR*28基因多态性与阿托伐他汀调脂疗效的关系。方法采用聚合酶链反应-直接测序法对以阿托伐他汀治疗(20 mg,qn)30 d的185名高脂血症患者进行基因分型,比较治疗前后各基因型间血三酰甘油(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)及高密度脂蛋白胆固醇(HDL-C)水平的差别。结果 POR*28的基因变异率为41.6%,治疗前各基因型间TG、TC、LDL-C和HDL-C的浓度均无显著差异;治疗30 d后,纯合子等位基因TT型携带组LDL-C浓度显著低于CC型携带组[(2.46±0.37)mmol·L-1vs.(2.69±0.49)mmol·L-1,P=0.019],各基因组TG、TC浓度均无显著差异(P>0.05);TT型携带组HDL-C浓度有升高的趋势(P>0.05)。结论 POR*28基因多态性与阿托伐他汀降低LDL-C浓度有关。AIM To investigate the associations between POR*28 polymorphisms and lipid-lowering response to atorvastatin. METHODS Genotypes were determined using polymerase chain reaction(PCR)-direct sequencing in 185 hyperlipidemic patients treated with atorvastatin 20 mg once daily for 30 days. Serum triglyceride( TG), total cholesterol( TC), low-density lipoprotein cholesterol( LDL-C), and high density lipoprotein cholesterol(HDL-C) levels were determined before and after the treatment. RESULTS Variant allele frequencies of POR*28 was 41.6%. There were no statistically significant differences in the levels of TG, TC,LDL-C and HDL-C before treatment among the POR*28 genotypes. After 30 days treatment, the level of LDLC after treated with atorvastatin was significantly decreased in subjects with homozygous for the T allele than forthe C allele of POR*28(( 2.46 ± 0.37) mmol·L-1vs.( 2.69 ± 0.49) mmol·L-1, P = 0.019). There were no significant differences in levels of TG and TC between the T and C alleles( P 〉 0.05). And the increased tendency was seen in the level of HDL-C(P 〉 0.05) in the T alleles. CONCLUSION POR*28 polymorphism is related to LDL-C lowering response of atorvastatin.
