摘  要:目的采用超高效液相色谱法(UPLC)测定人血清中的顺铂。方法色谱柱为Acquity UPLC BEH C18柱(100 mm×2.1 mm,1.7μm),流动相为75%甲醇,检测波长254 nm,流速1.0 m L·min^-1,柱温35℃。结果顺铂的标准曲线为:Y=3.1×10-3X+0.0685(r=0.9999),顺铂0.1~10μg·m L^-1与峰面积的线性良好;最低检测限为50 ng·m L^-1,日内、日间RSD分别小于5.0%、10.0%,提取回收率〉70.0%。结论所用方法快速、灵敏、准确、专属性强,适用于顺铂血药浓度的监测及药动学研究。OBJECTIVE To determine Cisplatin in patients serum by UPLC. METHODS UPLC analysis was performed on a Acquity UPLC BEH C18column( 100 mm × 2. 1 mm,1. 7 μm). The mobile phase consisted of methanol-water( 75∶25) with the flow rate at 1. 0 m L·min^-1. The column temperature was 35 ℃. The detection wavelength was set at 254 nm. RESULTS Cisplatin showed good linearity in the range of 0. 1-10. 0 μg·m L^-1,linear regression equation of Y = 3. 1 × 10-3X + 0. 0685( r = 0. 9999) and with the detectable limitation of 50 ng·m L-1. The intra-day and inter-day RSD were less than 10. 0% and 5. 0%,respectively and the average recoveries were higher than 70. 0%. CONCLUSION The method is rapid,sensitive,accurate and specific,and suitable for the blood drug concentration monitoring of Cisplatin in the clinic.
