摘 要:为探讨GDF5基因外显子2的SNPs以及与鸡胫骨长的关联性,以兴义矮脚鸡GDF5基因为研究对象,采用基因克隆技术得到外显子2序列,直接测序后检测SNPs,并与胫骨长进行相关性分析。结果表明,共检测到5个SNPs,前4个位点(C146A、C296T、G326A和C578G)均为两种基因型,第5位点(C704T)为三种基因型。基因型与胫骨长性状关联分析显示,C578G位点CC基因型个体胫骨长显著高于GC型个体(P<0.05),其余SNPs均差异不显著(P>0.05)。研究表明,GDF5基因可能是影响鸡胫骨长的主效基因或与主效基因相连锁,C578G位点有望成为胫骨长性状选择的分子遗传标记,为家禽的选育提供理论依据。To analyze the corelation between the SNPs of GDF5 gene excon 2and shin length in chicken,Xingyi Bantam were selected to obtain the sequence of excon 2through gene cloning and detect the SNPs after direct sequencing.The results showed that 5SNPs were detected in excon 2,the previous 4SNPs(C146A,C296 T,G326Aand C578G)were divided into 2genotypes,the fifth SNP was in 3genotypes.The study of relationships between genotypes and shin length revealed that shin length of individuals with genotype CC were significantly higher than those with genotype GC in C578G(P〈0.05),no significant corelation about other SNPs was found(P〉0.05).The result indicated that GDF5 gene may be a potential major gene or linked major gene which significantly affects shin length in chicken,and C578 Ga candidate molecular market of MAS,providing theoretical basis for the breeding of poultry.
