摘 要:目的:观察菊苣提取物对高果糖饮水诱导的高甘油三酯并高尿酸血症大鼠尿酸、甘油三酯、总胆固醇水平的影响,并从肠道菌群新的视角探讨菊苣提取物干预高甘油三酯并高尿酸血症模型的特点。方法:雄性SD大鼠按体重随机分为6组,即正常组,模型组,菊苣高剂量组,菊苣低剂量组,非诺贝特组,苯溴马隆组。正常组给予自来水,其余各组均给予10%果糖水,生化法检测血清尿酸、甘油三酯、总胆固醇指标,ERIC-PCR指纹图谱分析各组肠道菌群宏观结构变化。结果:与正常组相比,造模第7天,模型组血清TG水平显著升高;造模第14天,模型组血清TG水平显著升高,血清UA水平有升高趋势;造模第21天,大鼠血清TG、UA均显著升高,大鼠高甘油三酯并高尿酸血症模型塑造成功。与模型组相比,实验第14天,菊苣高剂量,菊苣低剂量,非诺贝特,苯溴马隆均显著降低血清TG水平;实验第21天,菊苣高剂量组显著降低血清TG、UA水平;菊苣低剂量组有降低血清TG和UA水平的趋势;非诺贝特组显著降低血清TG水平;苯溴马隆组显著降低血清UA水平。ERIC-PCR指纹图谱显示:与正常组相比,模型组肠道菌群结构发生了显著变化,而菊苣高、低剂量组对肠道菌群有干预作用。结论:菊苣提取物治疗高甘油三酯并高尿酸血症作用显著,并可改善紊乱的肠道菌群,说明菊苣提取物可能通过调节肠道菌群发挥治疗作用,具体机制有待于进一步深入研究。Objective:To investigate the effect of chicory extracts on serum UA,TG and TC levels of hypertrig-lyceride in hyperuricemia rats induced by high fructose water,to discuss the possible characteristics of chicory extracts from the respect of gut flora.Methods: Male SD rats were randomly divided into 6 groups,a normal group,a control group, chicory groups with high and low doses, a fenofibrate group and a benzbromarone group.The normal group was given distilled water,and other groups were given 10%fructose water.Biochem-ical technique was used to detect serum UA,TG and TC levels,and gut flora macrostructure was analyzed by ERIC-PCR fingerprint.Results:Compared with the normal group, TG level in the control group significantly increased on the 7th day;TG level increased significantly,and UA level had the trend to increase on the 14th day;TG and UA levels increased simultaneously on the 21st day,and it showed that the rat models of hyper-triglyceride with hyperuricemia were established successfully.Compared with the control group,in the chicory groups with high and low doses,in the fenofibrate group and in the benzbromarone group UA level decreased significantly on the 14th day;in the chicory group with high dose TG and UA levels significantly decreased,and the chicory group with low dose had the trend to decrease TG and UA levels;in the fenofibrate group TG level decreased significantly and in the benzbromarone group UA level decreased significantly.ERIC-PCR finger-print revealed that compared with the normal group, the gut flora structure in the control group changed signifi-cantly,and in the meanwhile chicory groups with high and low doses could interfere the disordered gut flora. Conclusion:The effect of chicory extracts on hypertriglyceride with hyperuricemia is significant.In addition, it can improve disordered gut flora.It reveals that chicory extracts may work by changing gut flora in rats,but the specific mechanism remains to be further researched.
