护心胶囊对冠心病阿司匹林抵抗大鼠TXB_2和6-Keto-PGF_(1α)的影响

摘　　要：目的:观察护心胶囊对冠心病阿司匹林抵抗(aspirin resistance,AR)大鼠血栓素B2(thromboxane B2,TXB2)、6酮前列腺素F1α(6 ketone prostaglandin F1α,6-Keto-PGF1α)的影响,探讨其对阿司匹林抵抗机制。方法:选用SD雄性大鼠75只,随机分为5组:正常组、模型组、护心胶囊低剂量组、护心胶囊中剂量组、护心胶囊高剂量组,每组15只。除正常组外,前10周其余各组造成冠心病阿司匹林抵抗大鼠模型。从第11周开始,护心胶囊低剂量组、中剂量组、高剂量组分别灌服1.8g/kg、3.6g/kg、7.2g/kg护心胶囊混悬液,正常组及模型组灌以等量生理盐水,均灌胃4周。取大鼠股动脉血,检测血浆中TXB2、6-Keto-PGF1α的含量。结果:模型组较正常组血浆中TXB2含量明显升高(P<0.01),而6-Keto-PGF1α的含量却显著降低(P<0.01),护心胶囊低、中、高剂量组相对模型组血浆TXB2含量均明显降低(P<0.01),血浆6-Keto-PGF1α的含量均升高(P<0.05或P<0.01)。结论:护心胶囊可降低冠心病阿司匹林抵抗大鼠血浆TXB2水平,升高血浆6-Keto-PGF1α水平,有抗血小板聚集的功效,改善阿司匹林抵抗现象。Objective：To discuss the mechanism of aspirin resistance by observing the effect of HuXin capsules on TXB2 and 6-Keto-PGF1αin CHD rats with aspirin resistance.Methods：75 SD male rats were randomly divided into 5 groups：a normal group, a model group, HuXin capsule low-dose, HuXin middle-dose and high-dose groups, with 20 rats in each group.Except the normal group,in the rest groups CHD rat models with aspirin resistance were induced in the first 10 weeks.From the 11th week,in HuXin capsule low-dose, middle-dose and high-dose groups rats took 1.8g/kg, 3.6g/kg and 7.2g/kg HuXin capsule suspension for 4 weeks,and the rats in the normal group and model group were instilled with the same amount of saline solu-tion for 4 weeks.Take the femoral arterial blood of rats to detect TXB2 and 6-Keto-PGF1αcontents in the plasma.Results：Plasma TXB2 content in the model group compared with that in the normal group was obvious-ly higher（P〈0.01）,and the content of 6-Keto-PGF1αwas significantly reduced（P〈0.01）.HuXin cap-sule low-dose,middle-dose and high-dose groups compared with the model group, plasma TXB2 content was significantly lower（P〈0.01）,but plasma 6-Keto-PGF1αcontent was elevated（P〈0.05 or P〈0.01）. Conclusion：HuXin capsules can reduce the plasma TXB2 level and elevate the plasma 6-Keto-PGF1αlevel of CHD rats with aspirin resistance.It has an effect on preventing platelet aggregation and improving the aspirin resistance.
