摘  要:目的观察神经生长因子(nerve growth factor,NGF)对糖尿病大鼠视网膜神经细胞凋亡的影响,研究NGF对糖尿病视网膜神经的保护作用。方法选择清洁级雄性2月龄SD大鼠60只,随机分成正常组、糖尿病组和NGF组,每组20只,后两组制作糖尿病模型。NGF组给予NGF腹腔注射(1000 U·kg-1),每天1次,共注射10 d,正常组和糖尿病组腹腔注射等体积生理盐水,3组分别在注药后4周末和8周末随机选取10只大鼠处死,取视网膜组织,做电镜标本观察视网膜超微结构,制备视网膜石蜡切片,采用TUNEL法进行凋亡细胞原位标记,行视网膜神经细胞凋亡计数,对所有数据进行统计学分析。结果电镜下正常组视网膜结构正常;糖尿病组膜盘局部模糊,线粒体肿胀,嵴消失,染色质边集;NGF组神经细胞无明显改变,仅见视杆细胞膜盘局部模糊不清,内核层和外核层染色质轻度密集,改变较糖尿病组轻。视网膜凋亡指数4周、8周时正常组分别为(3.30±1.52)%和(3.40±1.10)%,糖尿病组分别为(21.53±5.34)%和(31.40±10.15)%,NGF组分别为(12.36±5.26)%和(15.47±6.73)%,不同时间点3组间差异均有统计学意义(均为P=0.000)。结论 NGF能有效降低糖尿病大鼠视网膜神经细胞的凋亡,对糖尿病视网膜神经细胞有保护作用。Objective To study the effects of nerve growth factor(NGF) on ap- optosis in retinal nerve cells of rat with diabetes, and research the protective effect of NGF on retinal nerve cells with diabetes. Methods Sixty Male SD rats were randomly divided into negative control group, diabetic model control group and NGF treated group. Diabetes model was established by intraperitoneal injection of 60 mg~ kg-1 streptozotocin. 1000 U ~ kg-~ NGF was given by intraperitoneal injection one time per day for l0 consecutive days in the rats of NGF group ,and an equivalent amount of phys- iological saline was utilized at a same way in the animals of negative group and diabetic group. At 4 weeks and 8 weeks after diabetic induction, the eyeballs were randomly exe- cuted in l0 rats of each group for paraffm section and observed under optical micro- scope. Retina were observed under transmission electronic microscope, and TUNEL staining was used to observe neuron apoptosis in retina. ][~s~ Transmission elec- tronic microscope showed that in negative group, the structures of cells were normal;In diabetes group, the membrane disks were obscure and mitochondria were swollen, crest disappeared and chromatin were concentrated;In NGF group, no significant changes was found in nerve cell with integrited membranes, normal space around nuclear and endoplasmic reticulum arranged orderly;Changes with mildly swelling mitochondria, ob- scure cristae and chromatin were slighter than diabetic group. The apoptosis index at 4 weeks and 8 weeks in negative control group were(3.30 ± 1.52)% , (3.40 ± 1.10)% ,re- spectively,which in diabetic control group were(21.53±5.34)% , (31.40± 10. 15)% , respectively,NGF group were( 12.35 ± 5.25)% , ( 15.47 ± 5.73 )% ,respectively. There were significant differences at different time among three groups ( all P = 0.000).
