5%咪喹莫特乳膏对BALB/c小鼠皮肤组织和外周血VEGF mRNA的影响

摘　　要：目的研究5%咪喹莫特对BALB/c小鼠皮肤组织和外周血中血管内皮生长因子(VEGF mRNA)表达的影响,以探讨其诱导银屑病小鼠动物模型的可能作用机制。方法 5%咪喹莫特乳膏规律地涂抹于BALB/c小鼠背部皮肤,采用实时荧光定量PCR方法分别检测皮肤组织和外周血中VEGF mRNA的表达情况。结果 5%咪喹莫特乳膏涂抹小鼠背部皮肤5天后出现类似银屑病样皮损;实时荧光定量PCR结果示:涂药8d后,在皮肤组织和外周血中,实验组小鼠的VEGF mRNA表达量均高于正常对照组小鼠的VEGF mRNA表达量,且差异均有统计学意义(P均<0.05)。结论 5%咪喹莫特乳膏成功诱导银屑病动物模型的机理可能与其能上调皮肤组织和外周血VEGF mRNA的表达有关。Objective To study the effects of 5% imiquimod cream on the expression levels of VEGEF in skin and pe- ripheral blood of BALB/C mice, and to investigate the possible mechanisms by which 5% imiquimod cream induces psoriasis in mice. Methods Following topical applications of 5% imiquimod cream to the back of BALB/C mice, the expression levels of VEGEF mRNA in both skin and peripheral blood were measured u- sing RT-PCR. Results Topical applications of 5% imiquimod cream for 5 days induced psoriatic lesions in BALB/C mice. After 8 days of topical applications of 5% imiquimod cream, the levels of VEGEF mRNA in both skin and peripheral blood in imiquimod-treated mice were significantly higher than that in normal con- trois （P 〈 0.05 ）. Conclusion The mechanism by which 5% imiquimod cream induces psoriatic lesions could be associated with up-regulation of VEGEF mRNA in both skin and peripheral blood.
