摘  要:目的 注射用A型肉毒毒素中加入明胶作为稳定剂,建立并验证定量检测注射用A型肉毒毒素中明胶含量的方法.方法 通过苦味酸与明胶特异性作用,产生强的吸收,采用紫外-可见分光光度计用外标法来检测注射用A型肉毒毒素中的明胶含量.结果 通过一定浓度苦味酸与明胶作用,用分光光度计在520 nm处测定吸光度,检测限可以达到2.5 mg/L,检测范围可以达到10 ~ 100 mg/L.结论 该方法具有良好的特异性、准确度、精密度和灵敏度,对于测定注射用A型肉毒毒素中的明胶含量有较好的参考价值.Objective To establish a method in determination of gelatin content in botulinum toxin type A for injection, the gelatin is a stabilizing agent existing in the product. Methods Based on the specifical reaction between picric acid and ge- latin, it produces a strong absorption for wavelength, an external standard method was performed to determinate gelatin con- tent in botulinum toxin type A for injection by using an UV-visible spectrophotometer. Results After applying a certain concentration of pierie acid to react with gelatin, the absorbance was measured at a wavelength of 520 nm with the detection limit of 2.5 mg/L and the test range from 10 mg/L to 100 mg/L. Conclusions This method has high specificity,accuracy,preci- sion and sensitivity, resulting in good reference value in detecting gelatin content in botulinum toxin type A for injection.
