LY294002对糖尿病肾病大鼠肾组织MCP-1与Nephrin表达的影响

摘　　要：目的研究经静脉注射PI3K抑制剂LY294002对糖尿病肾病(DN)大鼠模型肾组织单核细胞趋化蛋白-1(MCP-1)与肾小球足细胞跨膜蛋白(Nephrin)表达的影响。方法成年健康清洁级SD雄性大鼠30只,随机分为假手术组(10)、模型组组(10)和PI3K抑制剂(LY294002)处理组。参照Anderson's方法制作糖尿病肾病大鼠模型。采用Western blot方法检测3组大鼠肾组织MCP-1与Nephrin蛋白的表达,RT-PCR方法检测三组大鼠肾组织MCP-1 m RNA与Nephrin m RNA的表达。结果模型组肾组织MCP-1蛋白和m RNA的表达量显著高于假手术组(P<0.05),而LY294002处理组明显低于模型组(<0.05)。模型组肾组织Nephrin蛋白和m RNA的表达量显著低于假手术组(<0.05),而LY294002处理组显著高于模型组(<0.05)。结论 PI3K抑制剂LY294002能够下调糖尿病肾病大鼠肾组织MCP-1的表达,上调Nephrin的表达。Objective To study the effect of LY294002 on the expressions of monocyte chemoattractant protein-l（MCP-1） and Nephrin in diabetic nephopathy rat kidney. Methods 30 healthy male rats were selected and randomly divided averagely into the sham group, the model group and LY294002 treated group. The diabetic nephropathy model was induced with Anderson＇s way. The expressions of MCP-1 and Nephrin were observed by western blot method; The expressions of MCP-1 mRNA and Nephrin mRNA were observed by RT-PCR method. Results The expression levels of MCP-1 protein and mRNA were increased significantly in model than in sham group, but decreased in LY294002 treated group than in mode（P〈0.05）l. The expression levels of Nephrin protein and mRNA were decreased significantly in model than in sham group, but increased in LY294002 treated group than in model（P〈0.05）. Conclusion LY294002 could down-regulate the expression of MCP-1 and up-regulate the expression of Nephrin in diabetic nephooathv rat kidney.
