摘  要:目的:探讨心肌球样细胞(CDCs)在心力衰竭治疗中的应用前景。方法 Wistar雄性大鼠24只,无菌下取大鼠心脏组织通过胶原酶消化,培养取得CDCs,采用体外培养连续传代(P1~P5),镜下观察可见CDCs团形成。采用流式细胞仪分析细胞表型,并连续观察CDCs的自我更新和心肌细胞分化能力,并用免疫组化检测CDCs中的肌钙蛋白(cTnT)的表达。结果CDCs体外呈指数生长并形成细胞团样结构,流式细胞仪检测显示:CD29、CD44、CD90和CD106为阳性,而CD34、CD45为阴性。CDCs表达了心肌细胞特异性的结构蛋白cTnT,并可在体外培养条件下产生自主搏动。结论 CDCs是具有心肌细胞分化潜能及临床应用前景的细胞供体。Objective To investigate the differentiation potential of cardiosphere- derived cel s ( CDCs ) after expansion in vitro . Methods CSCs were isolated by enzymatic digestion of cardiac issue from 24 Wistar rats and their immunological phenotypes were analyzed by using FACS and immunohistological staining . After expansion up to 5th passage, the ability of self- renewal and myogenic potential were analysed. Results The isolated cardiac stem cel s(CSCs) grew exponential y and exhibited a similar morphology to cardiosphere. FACS analysis revealed positive for CD29, CD44, CD90, CD106and negative for CD34, CD45. Smal fractions of CSCs were undergoing myogenic differentiation, evidenced by the expression of cardiac structural proteins (cTnT) and developing spontaneous contraction. Conclusion CDCs retain myogenic potential after in vitro expansion and thus may be used as donor cel s for cardiac plasticity.
