摘 要:目的探讨参苓白术散对脾虚湿困型溃疡性结肠炎(UC)大鼠结肠组织NF-κB p65蛋白表达及相关炎性因子的影响。方法将48只Wistar大鼠按照随机数字表分为正常组、模型组、柳氮磺吡啶组、参苓白术散组,每组12只,脾虚湿困型溃疡性结肠炎大鼠模型采用三硝基苯磺酸(trinitrobenzene sulfonic acid,TNBS)/乙醇灌肠结合环境与饮食干预法复制,运用免疫组织化学法检测并比较各组大鼠结肠组织NF-κB p65蛋白表达,ELISA法测定血清IL-6、IL-8、TNF-α水平的变化。结果模型组大鼠NF-κB p65蛋白表达及IL-6、IL-8、TNF-α水平明显高于正常组(P<0.05),参苓白术散组和柳氮磺吡啶组较模型组显著降低,差异有统计学意义(P<0.05),参苓白术散组与柳氮磺吡啶组比较差异无统计学意义(P>0.05)。结论参苓白术散可以明显改善UC大鼠结肠组织中NF-κB p65蛋白表达及血清IL-6、IL-8、TNF-α水平,促进肠黏膜的修复。Objective To investigate the effects of Shenlin Baizhu San ( Ginseng, Poria and Atractylodes Macrocephalae Powder) on the protein expression of NF-κB p65 in the colon tissue of rats with ulcerative colitis due to dampness retention and spleen deficiency, as well as the related inflammatory cytokines. Methods 48 Wistar rats were divided into four groups: normal group, model group, sulfasalazine group, and Shenlin Baizhu San group, with 12 rats in each group.The model rats were induced by environment and diet intervention combined with composite trinitrobenzene sulfonic acid and ethanol in enema.The protein expression of NF-κB p65 in the colon tissue of rats were detected with immunohisto-chemistry methods.Levels of IL-6,IL-8, and TNF-αwere measured with ELISA.Results The protein expression of NF-κB p65 and the serum level of IL-6, IL-8, and TNF-αin the model rats were significantly higher than those in normal group ( P〈0 .05 ) .The protein expression of NF-κB p65 and the serum level of IL-6, IL-8, and TNF-αin Shenlin Baizhu San and sulfasalazine groups significantly decreased, and the difference was significant compared with model group (P〈0.05).The difference between Shenlin Baizhu San and sulfasalazine groups showed no statistical significance ( P 〉0.05 ) . Conclusion Shenlin Baizhu San could significantly improve the protein expression of NF-κB p65 and the serum level of IL-6,IL-8, and TNF-αin the colon tissue of rats with ulcerative colitis, which could promote the repair of the intestinal mucosa.
