摘  要:目的建立高效毛细管电泳法快速检测猪肉和内脏中克伦特罗、沙丁胺醇和特布他林含量的方法。方法匀浆样品经丙酮溶液超声波萃取,萃取液经离心和过滤后用氮气吹干。用磷酸盐缓冲液溶解残渣,低温离心,取上清过滤后进样。电泳条件:石英毛细管(75/365μm,40/47 cm),缓冲液为2μg/ml季铵化纤维素负载的纳米金磷酸盐缓冲液(p H=10.5),电压为12 k V,检测波长为210 nm。结果 3种物质均在5 min内出峰。浓度为1μg/ml^250μg/ml时,3种物质的峰面积和浓度的线性关系良好,相关系数r分别为0.996 7、0.994 8和0.995 3。猪肉和猪肝中3种物质的加标回收率为87%~113%,精密度RSD为2.8%~6.5%。结论该方法具有快速、简便、准确度和精密度高等特点,适用于猪肉和猪肝中克伦特罗、沙丁胺醇和特布他林的快速检测。Objective To develop a new method for rapid determination of clenbuterol, salbutamol and terbutaline in pork and liver by capillary electrophoresis. Methods Acetone combined with ultrasonic extraction was used to elute β2 - agonists from samples. The eluant was evaporated by nitrogen and was redissolved in phosphate buffer, and then the supernatant was analyzed by CE after centrifugation and filtration. The capillary was fused silica capillary with id/od of 75/365μm and effective/total length of 40/47 cm. Running buffer was 2 μg/ml QC - Au NPs phosphate buffer. Separation voltage was 12 kV. Detection wavelength was set at 210 nm. Results The crest could be completed within 5 minutes. Good linearity was found within the range of 1 μg/ml - 250 Ixg/ml, and the r was 0.996 7, 0.994 8 and 0. 995 3 respectively. The recoveries of three β2 - agonists in pork and liver ranged from 87% to 113%, and the relative standard deviations of precision (RSDs) ranged 2.8% -6.5%. Conclusion This method is fast, simple, precise and it is feasible for the determination of clenbuterol, salbutamol and ter- butaline in pork and liver tissues.
