消冻灵乳膏对豚鼠瘙痒模型止痒作用的实验研究

摘　　要：目的:观察消冻灵乳膏对豚鼠致痒阈的影响,从而研究其止痒作用。方法:造模前24 h,在豚鼠右后脚背剃毛(约1 cm2),随后涂抹相应药物。实验当日用三棱针在各组剃毛处点刺,以轻度渗血为度,其后涂抹相应药物2次,剂量为0.1 g/100 g体重,每次间隔时间1 h。末次给药15 min后,在扣刺处涂抹0.01%磷酸组胺0.05 m L/只,其后每隔3 min依次涂抹0.02%、0.03%、0.04%、0.05%、0.1%、0.2%、0.3%的磷酸组胺0.05 m L/只,观察豚鼠反应,待回头舔舐右后脚,记录磷酸组胺总量(mg),即为致痒阈。结果:实验结果显示消冻灵乳膏低中高剂量组、阳性药物对照组致痒阈明显高于模型对照组(P<0.05),表明阳性药Benadryl凝胶及消冻灵乳膏各剂量均能提高致痒阈。结论:消冻灵乳膏能提高豚鼠致痒阈,具有一定的止痒作用。Objective： To explore the antipruritus action of Xiao Dong Ling Ru Gao by observing its effect onitch-threshold of guinea pig. Methods： The right rear foot of guinea pig was shaved for about 1 cm^2, and applied with Xiao Dong Ling Ru Gao 24 hours before inducing pruritus model. The shaved area was pricked into mild bleeding withthree-edged needle, and applied with relative creams of 0. 1 g/100 g · bw every 1 hour for twice. 15 minutes after the last application, the pricked area was applied 0. 05 mL histamine phosphate in different concentrations （ 0. 02%, 0. 03%, 0. 04%, 0.05%, 0. 1%, 0. 2%, and 0. 3% ） every 3 minutes. The total dose of histamine phosphate was recorded as the guinea pig turn round to lick the rear foot, which is the itch-threshold. Results： The results shown that the itch-threshold of medication administration groups with low, medial and high dose of Xiao Dong Ling Ru Gao and positive control group was higher than vehicle control group （P 〈 0. 05） . Conclusion ： Xiao Dong Ling Ru Gao can relieve itching manifested by improving the itch-threshold.
