Optimization of procyanidins extraction from pericarp of Litchi chinensis by response surface methodology

摘  要:为优化荔枝皮中原花青素的提取工艺,以粤西地区所产的黑叶荔枝果皮为材料,在单因素考察的基础上,根据Box-Behnken设计原理,采用Design-Expert 8.0设计软件,以原花青素得率为指标,进行响应面优化试验,并通过DPPH法评价提取物清除自由基的能力。结果表明,荔枝皮的最佳提取工艺为乙醇浓度68%、提取温度59℃、提取时间93 min、液料比20∶1,提取2次,在此条件下荔枝皮原花青素的得率为5.26%。优化后原花青素的得率有所提高,经纯化后提取物清除自由基的能力与Vc相当甚至更优。To optimize the conditions for extracting procyanidins from pericarp of Litchi chinensis, Heiye growing in western Guangdong was used as experimental material. Based on single factor test and the Box-Behnken design principles, using procyanidins yield as the index, a response surface optimization test was established by using Design- Expert8.0 design software. And the efficiency of antiradical of procyanidins extracted from pericarp of Litchi chinensis was determined by the DPPH assay. The results show that the optimized extraction conditions of procyanidins from pericarp of Litchi chinensis were obtained as follows: ethanol concentration was 68%, extraction temperature was 59℃, extraction time was 93 min, solid-liquid ratio was 20 : 1 and extracted twice. Under these conditions, the yield of proeyanidins was 5.26%. The yield of procyanidins increased after optimizing, and the ability of antiradical of the purified procyanidins was similar or even better than Vc.
