紫外诱变对侧耳菌生长、酶活性及纤维降解的影响

摘  要:研究旨在通过紫外诱变育种技术提高侧耳菌纤维降解能力,来提高其处理秸秆饲料的效率。试验以侧耳菌为出发菌株,进行紫外诱变,采用PDA培养基筛选生长速度较快的菌株,再经过含有87%秸秆的培养基进行复筛,选育高效纤维降解菌株。结果表明:以紫外线照射30 s为最佳诱变剂量,获得菌株P89,其木质素过氧化酶(Li P)和漆酶(Lac)活性分别为出发菌株的2.03倍和1.89倍,菌丝生长速度提高18.21%,木质素含量降低19.89%(P<0.05),中性洗涤纤维(NDF)和酸性洗涤纤维(ADF)含量分别降低5.28%(P<0.01)和5.16%(P<0.01)。菌株P89为本研究诱变筛选获得的开发秸秆资源优秀菌株。The aim of the research was to adopt ultraviolet ray mutation dealing with Pleurotus ostrea-tus to improve the ability of fiber degradation and efficiency of treat corn straw. In order to get betterstrain with high fiber degradation, Pleurotus ostreatus was performed by UV mutagenesis, comparedthe rote of mycelia growth in PDA medium for the preliminary screening and the second screeningwas processed in the medium containing 87% straw powder. Results indicate that determined 30 swas the optimal dose, then the best strain P89 was gained. Li P and Lac enzyme activity of P89 was2.03 times and 1.89 times of the control group, the rote of mycelial growth improved 18.21%, lignincontent declined 19.89%(P〈0.05), NDF content and ADF content declined 5.28%(P〈0.01) and 5.16%(P〈0.01), respectively. In conclusion, P89 was the best strain for exploiting straw resources in the re-search.
