摘 要:采用絮凝作为前处理,结合超滤的方法,以咖啡酸和黄芩苷的保留率、蛋白质去除率和水提液的澄清度为指标,优化蒲地蓝中药水提液除杂工艺.确定了本实验絮凝处理后的最佳超滤参数:三味水提液为隔膜泵工作频率20,Hz、操作压力1.0,MPa;黄芩水提液为隔膜泵工作频率20,Hz,操作压力0.6,MPa.经过此工艺处理,三味水提液的蛋白质去除率为81.11%,相对澄清率为91.17%,左右,咖啡酸保留率为75.09%;黄芩水提液的蛋白质去除率为79.39%,相对澄清率为95%,左右,黄芩苷保留率为40.65%,.结果表明:絮凝和超滤耦合工艺用于蒲地蓝中药水提液除杂,比单纯絮凝工艺可更有效地提高杂质去除率,保证药液澄清度和稳定性,但是黄芩苷的保留率较低.Using flocculation for pretreatment, combined with ultrafiltration method, the purification process for water- extraction solution of Pudilan was optimized, which justified proved by the retention rate of caffeic acid and baicalin, the protein removal rate and the clarity of the oral liquid. The experiment results show that the following ultrafiltration conditions should be realized after flocculation: the Sanwei water extract is extracted under 20 Hz diaphragm pump frequency and 1.0 MPa operating pressure, the Scutellariae water extract is extracted under 20 Hz diaphragm pump frequency and 0.6 MPa operating pressure, and then the effect of removing can be obviously improved. The protein removal rate of the Sanwei water extract was 81.11%, the relative clarity rate was 91.17%, and the retention rate of caffeic acid was 75.09%. As to the Scutellariae water extract, the protein removal rate was 79.39%, the relative clarity rate was 95%, and the retention rate of baicalin was 40.65%. As a result, the flocculation combined with ultrafiltration process for the water-extraction purification of Pudi- lan was more effective than only using flocculation process for ensuring the clarity and stability of the liquids. But the reten- tion rate of baicalin is low.
