生存素、环氧合酶-2在多囊卵巢综合征大鼠子宫内膜组织中的表达及二甲双胍对其影响的研究

摘　　要：目的观察多囊卵巢综合征(PCOS)大鼠子宫内膜组织中生存素(Survivin)、环氧合酶-2(COX-2)的表达情况,探讨二甲双胍对PCOS内膜病变的治疗作用机制。方法造模成功后,将模型大鼠分为干预组(15只)和PCOS组(15只)。干预组给予二甲双胍灌胃,PCOS组给予生理盐水灌胃。同时另外选取15只健康SD大鼠作为对照组。分别对3组大鼠进行口服葡萄糖耐量试验(OGTT)及血清空腹胰岛素(FIN)水平的测定。同时观察各组大鼠子宫内膜组织学改变和Survivin,COX-2的表达情况。结果 1应用米非司酮第4天大鼠出现阴道上皮细胞持续角化。2PCOS组空腹血糖(FPG)、服糖后1h血糖(1hPPG)、服糖后2h血糖(2hPPG)及血清FIN水平明显高于对照组(P<0.05);干预组FPG,1hPPG,2hPPG及血清FIN水平明显低于PCOS组(P<0.05)。3PCOS组大鼠子宫内膜Survivin及COX-2的表达均高于对照组(P<0.05);干预组大鼠子宫内膜Survivin及COX-2的表达明显低于PCOS组(P<0.05)。结论 PCOS大鼠子宫内膜的病变与Survivin及COX-2在子宫内膜组织中的表达关系密切;二甲双胍对PCOS有治疗效果。Objective To investigate the expressions of Survivin and cyclooxygenase-2（COX-2）in the endometrium of rats with polycystic ovary syndrome（PCOS）and explore the mechanism of the treatment of PCOS endometrial lesions.Method After successful modeling,the model rats were divided into an intervention group（n=15）and a PCOS group（n=15）.The intervention group were given metformin intragastric administration while the PCOS group were received normal saline.Meanwhile,15 healthy Sprague Dawley（SD）rats were selected as the control group.All three groups of rats were taken their oral glucose tolerance test（OGTT）and detected their levels of serum fasting insulin（FIN）.The rats were observed their endometrial histological changes and expressions of Survivin and COX-2.Results ① The vaginal epithelial cells showed continued keratosis on the fourth day after rats took Mifepristone.②In the PCOS group,the levels of fasting plasma glucose（FPG）,1hour plasma glucose after oral glucose（1hPPG）,2hour plasma glucose after oral glucose（2hPPG）and serum FIN were significantly higher than those of the control group（P〈0.05）;the levels of FPG,1hPPG,2h PPG and serum FIN in the intervention group were significantly lower than those of the PCOS group（P〈0.05）.③ The expressions of Survivin in the endometrial glandular epithelium and COX-2in rats of the PCOS group were higher than the control group（P〈0.05）.The expressions of Survivin and COX-2of the intervention group were lower than the PCOS（P〈0.05）.Conclusion PCOS endometrial lesions in rats are closely related to expressions of Survivin and COX-2in their endometrial tissues and metformin has a therapeutic effect on PCOS.
