健骨伸筋汤对大鼠膝关节炎关节软骨病理改变及关节液IL-1β及NO含量的影响

摘　　要：目的观察健骨伸筋汤对大鼠膝关节炎关节软骨病理改变及关节液IL-1β及NO含量的影响。方法 45只雌性SD大鼠随机分为5组,每组9只。通过改良的Hulth法制成肾虚型大鼠的膝骨性关节炎模型,2周后,对空白对照组、模型组生理盐水灌胃;治疗组分低、高浓度进行中药灌胃,于灌胃后第7周、13周各组随机挑选一半观察大鼠膝关节炎关节软骨病理改变及关节液IL-1β及NO含量的变化。结果造模后大鼠膝关节软骨评分远高于空白对照组,软骨破坏严重,差异有统计学意义(P<0.01);经治疗后,评分均有所降低,IL-1β和N O含量有所改善,说明治疗对膝关节软骨有修复作用,健骨伸筋汤低剂量组、高剂量组与壮骨关节丸组比较,差异有统计学意义(P<0.01)。结论健骨伸筋汤可降低IL-1β和NO含量,有一定的关节软骨保护作用,可一定程度地延缓膝骨关节炎动物模型软骨的退变。Objective To explore the effects of Jiangu Shenjin decoction in changes of the cartilage and IL-1β, NO of knee osteoarthritis of rats. Methods： KOA experimental animal mode was applied in this experiment, 45 female SD rats were randomly divided into 5 groups,9rats in each group. Rats were got knee osteoarthritis by hulth. Two weeks later, the nomal group and model group were fed Physiological saline.The treatment groups of low concentration Jiangu Shenjin decoction and high concentration Jiangu Shenjin decoction were fed low or high concentration Jiangu Shenjin decoction. Observing the changes of the cartilage and IL-1β, NO of knee osteoarthritis of rats in seven week and thirteen week. Results The changes of the cartilage of the rats by hulth was worse obvious than that of the normal rats, the difference was statistically significant（P〈0.05）. After treatment, the changes of the cartilage and IL-1β, NO were improved. It proved that cartilage of osteoarthritis rats was repaired. Groups of low concentration Jiangu Shenjin decoction and high concentration Jiangu Shenjin decoction were better than those of the the matched group, the difference was statistically significant（ P〈0.05）. Conclusion Jiangu Shenjin decoction can decrease the content of the IL-1β, NO, and protect cartilage of knees and put off the degeneration of cartilage.
