摘  要:目的建立测定小鼠组织中氟桂利嗪的HPLC-UV方法,研究单独灌胃给予盐酸氟桂利嗪和联合盐酸普萘洛尔灌胃后小鼠组织中氟桂利嗪的分布。方法 30只健康雄性昆明种小鼠随机分成2组,一组灌胃给予盐酸氟桂利嗪(14 mg·kg-1),另一组灌胃给予盐酸氟桂利嗪(14 mg·kg-1)和盐酸普萘洛尔(14mg·kg-1),用HPLC-UV法测定组织中氟桂利嗪的浓度。结果小鼠各组织中氟桂利嗪浓度在按2种给药方案给药1.0 h后达到最大,单独给药组和联合给药组小鼠各组织中氟桂利嗪的最大浓度:胃分别为(14 075±3 948)和(17 521±3 377)ng·g-1,小肠分别为(6 712±3 646)和(9 136±7 022)ng·g-1,肾分别为(1 054±423)和(1 667±1 123)ng·g-1,脑分别为(1 018±390)和(1 636±643)ng·g-1,肝分别为(1 565±482)和(3 293±1 119)ng·g-1。2组数据经统计学比较,给药1.0 h后肝中药物浓度差异有统计学意义(P<0.05)。结论 2组小鼠中氟桂利嗪在胃、小肠中浓度明显大于肝、肾、脑中的浓度。盐酸普萘洛尔可使盐酸氟桂利嗪在被测小鼠肝中的分布量增大,对药物组织分布有一定影响。Objective To establish an HPLC-UV method to determine flunarizine in mouse tissue, and the differences in the distribution of single administration of flunarizine hydrochloride and combination with propranolol hydrochloride in mouse tissue. Methods Totally 30 healthy male mice were divided into 2 groups: one group were given flunarizine hydrochloride (14 mg kg - 1) by intragastric administration gavage, while the other group were given fltmarizine hydrochloride (14 mg kg - 1) and propranolol hydrochloride (14 mg kg - 1). The concentration of flunarizine in the tissue was detected by HPLC-UV. Results The highest concentration of flunarizine in the tissue occurred 1.0 h in both methods of administration. The highest concentrations of flunarizine after single administration and combined administration in different parts of the body were as follows: the stomach (14 075 ± 3 948) and (17 521± 3 377) ng g -1, the small intestine (6 712 ± 3 646) and (9 136 ± 7 022) ng g- 1, the kidney (1 054 ± 423) and (1 667±1 123) ng g - 1, the brain (1 018±390) and (1 636±643) ng g - 1, and the liver (1 565±482) and (3 293±1 119) ng g - 1. There was significant difference in the concentration in the liver 1.0 h after the administration in both groups (P 〈 0.05). Conclusion The concentration of flunarizine in the stomach and the small intestine is significantly higher than that in the liver, the kidney and the brain. Combined administration with propranolol hydroehloride can increase the concentration of flunarizine in the liver, which has effect on the distribution of flunarizine hydrochloride in mouse tissue.
