摘 要:目的探讨瑞舒伐他汀对血脂正常原发性高血压患者氧化应激的影响。方法100例血脂正常的原发性高血压患者,随机分为安慰剂组50例(对照组)、瑞舒伐他汀治疗组50例(治疗组),疗程12周。2组患者在常规降压治疗的基础上分别每晚睡前加服安慰剂1片、瑞舒伐他汀10mg。检测所有患者治疗前后血脂(TC、TG、HDL、LDL)、血浆中8-羟基脱氧鸟苷酸(8-OHdG)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GPx)、丙二醛(MDA)水平。结果治疗前2组的血脂、8-OHdG、MDA、SOD、GPx差异无统计学意义(P>0.05),对照组治疗前后上述指标变化差异无统计学意义(P>0.05);治疗组治疗后患者血浆中8-OHdG、MDA水平下降(P<0.05),SOD、GPx水平上升(P<0.05),血清TC、LDL水平显著下降(P<0.05),HDL水平显著升高(P<0.05)。治疗后,除TG外,治疗组各指标均较对照组变化明显(P<0.05)。治疗组治疗前后8-OHdG、MDA与收缩压(SBP)正相关(P<0.05),SOD、GPx与SBP负相关(P<0.05),而各氧化指标与血脂无明显相关性(P>0.05)。结论氧化应激与高血压病情进展有一定相关性,而瑞舒伐他汀能抑制原发性高血压患者氧化应激。Objective To determine the effect of rosuvastatin on oxidative stress in essential hypertension patients without hyperlipemia. Methods Totally 100 essential hypertension patients without hyperlipemia were randomly divided into 2 groups: a control group (placebo, n = 50) and a treatment group (rosuvastatin 10 mg d - 1, n = 50). Serum of total cholesterol (TC), triglyceride (TG), high density lipoprotein (HDL), low density lipoprotein (LDL), 8-hydroxy-2-deoxyguanosine (8-OHdG), glutathioneperoxidas (GPx), malondialdehyde (MDA) and superoxidedismutase (SOD) were measured before and after 12 week treatment in the 2 groups. Results The serum levels of lipid, 8-OHdG, MDA, SOD, and GPx were not different in both groups before the treatment (P 〉 0.05). There was no significant change before and after the placebo treatment in the control group (P 〉 0.05). The serum levels of TC, LDL, 8-OHdG and MDA were decreased, while HDL, SOD and GPx levels were improved in the treatment group after the 12 week treatment (P 〈 0.05). The serum lipid did not change in all 3 groups (P 〉 0.05). Except for TG, the serum levels of 8-OHdG, TC, LDL and MDA were decreased, while HDL, SOD and GPx were increased in the 2 treatment groups compared with those in the control group after the treatment (P 〈 0.05). Systolic pressure (SBP) was positively associated with 8-OHdG and MDA and negatively associated with SOD and GPx (P 〈 0.05). There was no correlation between the oxidative stress parameters and lipid (P 〉 0.05). Conclusion There is correlation between oxidative stress and hypertension. Rosuvastatin dose dependently inhibits the oxidative stress in essential hypertension patients without hyperlipemia.
