扎冲十三味中矿物药对脑缺血保护作用初探

摘　　要：目的观察扎冲十三味中矿物药对大鼠脑缺血的保护作用并探讨其可能作用机理。方法采用拴线法复制大鼠脑缺血模型,将实验动物分成假手术组、脑缺血模型组和预治疗组(分别用扎冲十三味全方及其中矿物药按大、中、小剂量预先治疗27 d)。记录各组大鼠神经功能评分,计算脑梗死指数并用ELISA法检测血清炎症细胞因子IL-1、IL-6、TNF-α、IL-8及MCP-1水平。结果与脑缺血模型组相比,扎冲十三味矿物药大、中、小剂量组都不能影响脑缺血大鼠神经功能评分、脑梗死指数。模型组血清IL-1、IL-6、TNF-α、IL-8及MCP-1水平显著升高,矿物药大、中剂量均能不同程度降低上述血清因子水平(P<0.05);矿物药小剂量对脑缺血大鼠上述因子血清水平无显著影响(P>0.05)。结论扎冲十三味中矿物药大剂量、中等剂量应用均能明显抑制大鼠脑缺血过程中部分血清炎症因子水平,是扎冲十三味保护脑缺血损伤的重要组成成分,在方剂中可能与其他组分协同作用发挥药效。[Objective] To observe the protective effects of Garidi-13 mineral fraction on rats with cerebral ischemia and explore the possible mechanism. [ Methods ] Rats were randomly divided into sham operation group, model group, Garidi-13 group （0.24 g/kg）, large dose of minerals group （0.52 g/kg）, medium dose of minerals group （0.26 g/kg） and small dose of minerals group （0.13 g/kg）. Drug treatment groups were continuously administrated with the drugs by garage daily for 27 days, the rat model of focal cerebral ischemia was established. Neural function score, cerebral infarction index and serum levels of IL-1, IL-6, TNF-ot, IL-8 and MCP-1 were measured. [Results] Compared with the model group, the serum levels of IL-1, IL-6, TNF-ct, IL-8 and MCP-1 of the large and medium dose of minerals groups decreased significantly （P 〈 0.05）, while those were not decreased by the treatment with small dose of minerals（P 〉 0.05）. [ Conclusions ] Large and medium doses of mineral fraction could decrease the serum levels of IL-1, IL-6, TNF-α, IL-8 and MCP-1 during cerebral ischemia. It is an important moity of Garidi-13 in protecting cerebral ischemia injury, may have protective effect on cerebral ischemia with the drug combination.
