摘  要:目的:研究诱导型-氧化氮合酶(i NOS)和c GMP依赖性蛋白激酶1(PKG-1)在白藜芦醇(RES)防治肺动脉高压(PAH)中的分子机制。方法:大鼠随机分为对照组(Ct),治疗(RES)组和安慰剂(M),采用IPP软件测量分析肺小微动脉肥厚指数;免疫组化检测3组肺组织i NOS和PKG-1的表达。结果:m PAP在RES和M组间差异有统计学意义(P<0.05),右室肥厚指数、动脉肥厚指数RES和M组间差异明显(P<0.01);i NOS和PKG-1检测结果显示RES与M组间差异有统计学意义(P<0.001)。结论 :RES降低肺动脉压力,改善肺动脉结构,逆转肺动脉损伤和增生肥厚,与抑制i NOS的生成,增加PKG-1的表达关系密切。Objective To study the role of iNOS and PKG-1 inprevention and treatment of pulmonary artery hypertension (PAH) by resveratrol. Methods SD ratswererandomlydividedintocontrol, MCT injection andresveratrol (RES) treatedgroups. IPP software was used to count and measure micro arterial vascular hypertrophy. Western Blotand immunohistochemistrywere used to detect the expression of iNOS and PKG-1, and protein expression, then they were analyzed by IPP. Results Difference of mPAP, micro artery wall area and thickness and RV/(LV+S) between RES and M group were significantly different (P〈0.01). The depression of iNOS and activation of PKG-1 in RES group was significant (P〈0.001). Conclusion Resveratrol can preventPAH by inhibiting the expression of iNOS and increasing the PKG-1 expression.
