摘  要:目的 探讨一重与三重脑震荡(cerebral concussion,CC)后不同时期强迫游泳实验(forced swimming test,FST)抑郁样行为结构变化特点.方法 成年SD大鼠24只,用自制单摆闭合式脑损伤机械打击装置撞击大鼠额顶部,建立一重和三重CC大鼠模型,符合损伤判断标准的损伤大鼠被随机分为一次性脑震荡PCC组、及打击间隔分别为24 h与48 h的三重脑震荡3MCC 24 h组和3MCC 48 h组,另设一正常对照组,每组8只大鼠.应用FST行为学实验在伤后第3天、7天、15天、30天四个时间点进行大鼠损伤后抑郁样行为结构变化检测.结果 (1)与对照组相比,各损伤组大鼠的爬壁行为频数显著下降,均差异有统计学意义(P<0.05),其中各损伤组下降最大的时间点是:PCC组在伤后30 d为(6.00±3.30)次,3MCC48 h组在伤后7d为(5.75±3.01)次,3MCC 24 h组在伤后7d为(3.25±1.28)次,且爬壁行为的频数按PCC组>3MCC 48 h组>3MCC 24 h组依次递减;(2)与对照组相比,3MCC 24 h和3MCC 48 h组大鼠的静止不动行为频数均明显上升,均差异有统计学意义(P<0.05),其中各损伤组上升最大的时间点是:3MCC 48 h组在伤后7d为(19.00±2.83)次,3MCC 24 h组在伤后7d为(22.00±4.04)次,PCC组仅在伤后第30天差异有统计学意义,即(16.75±3.62)次,静止不动行为上升程度依次为PCC组<3MCC 48 h组<3MCC 24 h组.(3)游泳行为在各组间比较均差异无统计学意义(P>0.05).结论 一重与三重脑震荡大鼠损伤后出现不同程度的抑郁样行为,且三重CC重于一重CC、打击间隔短的重于打击间隔长的三重CC.Objective To observe the changes of depress-like behavior characteristics in different period of the forced swimming test(FST) after pure cerebral concussion(PCC) and multiple cerebral concussion(3MCC) model rats.Methods 24 adult healthy male SD rats were chosen.A brain concussion model was produced in rats using a simple pendulum device,according to the impacting times (one or three times) and impacting interval (24 h or 48 h).The rats were randomly assigned to the one time PCC group,triple MCC 24 h group(3MCC 24 h) and triple MCC 48 h group(3MCC 48 h),and one normal control group was used(8 rats in each group).FST was used to assess the change of depress-like behavior characteristics in rats.Results The following depress-like behavior changes of FST (climbing;swimming;motionless) were observed at the 3rd,7th,15th,30th day after injury were as follows:(1) Compared with the normal control group,the decline of "climbing" behavior frequency had statistically significant (P<0.05)in each injury group,the maximum decreased time in each injury group:(6.00 ±3.30)times after 30 d injury in PCC group,(3.25±1.28) times after 7 d injury in 3MCC 24 h group,(5.75± 3.01)times after 7 d injury in 3MCC 48 h group.Among all three injuried groups,the characteristic of "climbing" behavior had an decrease progressively as PCC group<3MCC 48 h group<3MCC 24 h group.(2)Compared with the normal control group,the increase of "motionless" behavior frequency had statistically significant(P<0.05)in both 3MCC injuried group at all four test points post-injury,the maximum increased time in each injury group:(19.00±2.83) times after 7 d injury in 3MCC 48 h group,(22.00±4.04) times after 7 d injury in 3MCC 24h group.But the PCC group had statistically significant (P< 0.05) only at the 30th day after injury,it was (16.75± 3.62) times.Among three injury groups,the characteristic of "motionless" behavior had an increase progressive
