摘 要:目的考察影响载蛋白聚乳酸聚乙醇酸(PLGA)微球形态和药物分布的因素。方法以异硫氰酸荧光素-牛血清白蛋白为模型药物,采用改良复乳化法制备载蛋白的PLGA微球,考察PLGA浓度和粘度,制备初乳和复乳时的搅拌转速对微球形态及药物分布的影响。以扫描电子电镜观察微球表面形态,激光共聚焦显微镜考察微球内部的药物分布。结果 PLGA浓度和粘度、制备初乳时搅拌转速的增加,均使药物在微球内分布均匀性增加。随着初乳搅拌转速的增加,微球表面孔洞数目增加,而复乳搅拌转速对微球形态和药物分布无明显影响。结论 PLGA的浓度和粘度以及初乳搅拌转速对微球形态和药物分布的影响较为显著。OBJECTIVE To investigate the influence of different parameters on the morphology and drug distribution of proteinloaded PLGA microspheres. METHODS The microspheres were prepared using modified double emulsion method. The effects of PLGA concentration and viscosity,stirring speed of primary and double emulsion on the morphology and drug distribution of PLGA microspheres loaded Fluorescein isothiocyanate labeled bovine serum albumin were investigated. Surface morphology was analyzed by scanning electron microscopy,and drug distribution was studied by confocal laser scanning microscopy. RESULTS Increasing the PLGA concentration and viscosity,and stirring speed of primary emulsion led to increase homogeneity of drug distribution. The number of the pores on the surface of the microspheres increased when the homogenization speed of primary emulsion increased. But different stirring speed of the double emulsion caused no significant effect on the morphology and drug distribution. CONCLUSION PLGA concentration and viscosity,and stirring speed of primary emulsion have significant effect on the morphology and drug distribution of the microspheres.
