摘 要:目的通过观察人肝7702细胞暴露于辛基酚(OP)后5-羟色胺(5-HT)转运体SERT、5-HT3b受体表达以及5-HT水平的变化,探讨OP对5-HT转运通路和受体结合通路的影响。方法实验分5个组,分别为OP高(5.0μg/L)、中(1.0μg/L)、低(0.2μg/L)剂量组、盐酸氟西汀组和空白对照组,体外细胞培养48 h,采用ELISA试剂盒检测细胞上清液中5-HT含量的变化,运用实时荧光定量PCR法检测转运体SERT及5-HT3b受体的表达。结果中、高剂量组细胞上清液中5-HT含量高于空白对照组(P<0.05),细胞中5-HT3b受体的表达降低(P<0.05);低、中、高剂量组细胞中SERT的表达均低于空白对照组(P<0.05);与盐酸氟西汀组检测结果一致。结论 OP可引起5-HT再摄取障碍,同时减少5-HT3b受体的表达,对机体5-HT代谢网络的转运通路及受体结合通路产生毒效应作用。[Objective] To explore the influence of Octylphenol (OP) on 5-hydroxytryptamine (5-HT) re- ceptor 5-HT3b and serotonin transporter (SERT) in human liver (HL) 7702 cells by means of detecting the level of 5-HT and observing expression levels of SERT and 5-HT3b in these cells. [Methods] Five groups were set for the study. Fluoxetine was for positive control, meanwhile other cultured cells were treated with different doses of OP [OP high (5 p.g/L), middle (1 I^g/L), low (0.2 p^g/L)] and the blank control group with in vitro cell culture for 48 h. The content of 5-HT in HL-7702 cells supernatant was detected by enzyme linked immunosorbent assay (ELISA). The expressions of SERT mRNA and 5-HT3b mRNA in HL-7702 cells were detected by real-time fluorescent quantitative PCR. [ Results ] The content of 5-HT in 1 Ixg/L OP and 5 Ixg/L OP groups was significantly higher than that in the blank control group (P〈 0.01), but the 5-HT3b levels were significantly decreased in the three OP groups (P 〈 0.05). The expressions of SERT in the OP groups signifi- cantly decreased compared to the blank control group (P 〈 0.05), the result was consistent with that of the flu- oxetine group. [ Conclusion] OP exposure causes the disorders of reuptake and binding with receptors of 5- HT through inhibiting SERT and 5-HT3b, so produces combined toxic effect in the transport pathway and re- ceptor binding pathway.
