摘  要:目的观察N-乙酰半胱氨酸(NAC)对模拟失重状态下大鼠肺损伤的影响。方法采用大鼠尾吊法建立模拟失重大鼠模型。清洁级Wistar大鼠24只,随机分为A组、B组和C组,每组各8只。从实验第1天开始B组每天给予N-乙酰半胱氨酸300 mg/kg药物灌胃,A组、C组予以等量无菌注射用水灌胃。第2天A、B两组进行持续尾吊建立模拟失重模型,C组大鼠于笼内自由活动,7天后将所有大鼠处死取材,测定血中性粒细胞表面CD11b/c、中性粒细胞活性氧浓度,白介素-10(IL-10)、白介素-6(IL-6)及肿瘤坏死因子-α(TNF-α)含量。取右肺上叶组织,切片行HE染色,观察肺组织结构变化。结果 A组显微镜下肺泡大小不一致,肺泡间隔水肿增宽,血管扩张充血,间质内可见中性粒细胞、淋巴细胞浸润。B组肺组织亦出现上述改变,但程度较A组轻,C组未见明显上述改变。各组间CD11b/c、活性氧(ROS)、TNF-α、IL-10、IL-6比较差异均有统计学意义(P<0.05,P<0.01)。结论 N-乙酰半胱氨酸能够减轻模拟失重状态下大鼠炎性反应和肺组织破坏,其抗氧化应激作用可能对肺组织损伤有一定保护作用。Objective To observe the effect of N-acetylcysteine (NAC) on pulmonary injury under simulated weightlessness in rats. Methods Tail-suspension method was used to establish the models of weightless simulation in rats. A total of 24 clean Wistar rats were randomly divided into group A, B and C (n = 8 for each group). Group B was treated with NAC (300 mg/kg) by intragastric administration every day from the first day, while group A and C were treated with same volume of sterile water via intragastric injection. The models of weightless simulation were established in group A and B by tail-suspension method after the rats had been administrated for 2 d, while the rats in group C were freerunning in the cage. The rats were killed at the end of the 7^th d. The levels of superficial CD11b/e of neutrophilic granuloeyte, reactive oxygen species (ROS) concentration of neutrophilic granuloeyte, contents of IL (interleukin)-10, IL-6 and TNF ( tumor necrosis factor) -α in the blood samples were detected. The superior lobe tissues of right lung were given hematoxylin and eosin stain ( HE stain), and the pathological changes of lung tissues in slices were observed. Results In group A, under microscope, the sizes of alveoli were discrepant, the alveolar septum became wide with edema, and blood vessels were dilated and congested, and neutrophils and infiltrated lymphoeytes were found in the interstitial. Similar changes of group A were found in group B, but the degrees were less than those in group A, while there were no obviously similar changes in group C. There were statistically significant differences in CD11b/c, ROS concentration, TNF- α, IL-10 and IL-6 (P 〈 0. 05, P 〈 0.01 ). Conclusion NAC can alleviate inflammation and lung damages of rats under simulated weightlessness, and the antioxidation effect of NAC may play a role in protection of lung injury under weightless condition.
