摘 要:目的探讨补肾与活血序贯干预对月经模型小鼠子宫内膜修复的影响及可能作用机制。方法将140只雌性C57BL/6J小鼠随机分为假手术组(40只)、模型组(50只)、序贯干预组(50只),模型组和序贯干预组采用切除小鼠双侧卵巢后行激素序贯处理复制小鼠月经模型。序贯干预组在激素序贯处理的同时序贯给予补肾方及活血方,每天相应药物2 ml/100 g灌胃,共计给药14天。模型组、假手术组灌胃等量蒸馏水。在孕酮撤退后24 h、32 h、40 h、48 h、72 h对各组小鼠诱导侧(右侧)子宫角进行形态学观察及评分,并检测各组小鼠子宫Cxcl 2蛋白的表达。结果与假手术组比较,模型组小鼠子宫内膜形态学评分在24 h、32 h、40 h、48 h显著降低(P<0.05);与模型组比较,序贯干预组小鼠子宫内膜形态学评分在24 h、32 h、40 h、48 h显著升高(P<0.05)。与假手术组比较,模型组Cxcl 2蛋白表达在各个时间点均升高(P<0.05);与模型组比较,序贯干预组Cxcl 2蛋白表达在各个时间点均升高(P<0.05)。结论补肾与活血序贯干预可促进月经模型小鼠子宫内膜修复,其作用机制可能与调控Cxcl 2蛋白表达有关。Objective To explore the effects and possible mechanism of sequential intervention of kidneyreinforcing and blood-activating on endometrium regeneration of menstruation model mice. Methods Totally 140 female C57BL /6J mice were randomly divided into sham-operated group( n = 40),model group( n = 50),sequential interventional group( n = 50). Model group and sequential interventional group adopted hormone sequential processing after removing bilateral ovaries of mice to copy menstruation model of mice. Sequential interventional group was given kidney-reinforcing decoction and blood-activating decoction sequentially while adopting hormone sequential processing,perfused with corresponding medicine 2 ml /100 g,for 14 days. Model group and sham-operated group were perfused with equivalent distilled water. Conducted morphological observation and scoring on guiding lateral( right) uterine horn of mice in each group 24 h,32 h,40 h,48 h,72 h after the withdrawal of progesterone,and detected the expression of Cxcl 2 protein of mice uteri. Results The morpholigical score of mice endometrium in model group decreased remarkably at 24 h,32 h,40 h,48 h compared with that of sham-operated group( P〈0. 05); The morpholigical score of mice endometrium in sequential interventional group increased remarkably at 24 h,32 h,40 h,48 h compared with that of model group( P〈0. 05). The expression of Cxcl 2 protein of model group all increased at any time point compared with that of sham-operated group( P〈0. 05); The expression of Cxcl 2 protein of sequential interventional group all increased at any time point compared with that of model group( P〈0. 05). Conclusion Sequential intervention of kidney-reinforcing and blood-activating can improve endometrium regeneration of menstruation model mice,and the underlying mechanism may be related to regulating the expression of Cxcl 2 protein.
