摘  要:目的探讨渗透压法检测冰冻解冻去甘油红细胞甘油残留量的可行性。方法用生理盐水、氯化钠-腺嘌呤-葡萄糖-甘露醇-磷酸二氢钠(MAP)液、血浆等不同介质溶液制备成含不同甘油浓度的洗涤红细胞,以模拟冰冻解冻去甘油红细胞,用渗透压仪测定其渗透压值,进行线性回归处理与相关系数的计算;并进行溶血干扰试验及重复性试验。结果渗透压值与甘油含量之间有很好的线性关系,但相同浓度的甘油在不同介质溶液(生理盐水、MAP液、血浆)悬浮的洗涤红细胞中的渗透压值有差异,生理盐水、MAP液、血浆悬浮的甘油浓度为10 g/L的洗涤红细胞的渗透压值分别为434、467、461 mmol/kg。游离血红蛋白在1.466-7.713 g/L时,渗透压测定值的差异无统计学意义。结论渗透压与甘油含量之间有良好的线性关系,溶血对渗透压测定影响小,可用于冰冻解冻去甘油红细胞的甘油残留量检测,且操作简单、快捷,但测定时应考虑制品的悬浮介质。Objective To investigate the feasibility of detecting residual glycerol in frozen thawed deglycerolized red blood cells(RBCs) using osmotic pressure method. Methods Washed RBCs with different glycerol concentrations were prepared by different mediums (saline, MAP additive solution and plasma) to simulate frozen thawed deglyccrolized RBCs. Then the osmotic pressure was measured by osmometer. The linear regression was used for data analysis and the correlation coefficient was calculated. Hemolytic interference test and repeatability test were done. Results There was a good linear relationship between the osmotic pressure and the concentration of glycerol. However, the osmotic pressure of washed RBCs was altered in different mediums (physiological saline, MAP solution and plasma). The osmotic pressures of washed RBCs with 10 g/L glycerol suspended in saline, MAP and plasma were 434 mmol/kg, 467 mmol/kg and 461 mmol/kg respectively. 1.466-7.713 g/L of free hemoglobin didn't affect the results. Conclusion There is a good linear relationship between the osmotic pressure and the concentration of glycerol. Hemolysis has no significant influence on determination of osmotic pressure. Osmotic pressure method is suitable for detecting residual glycerol in frozen thawed deglycerolized RBCs, with the advantages of easy and fast. However, the effect of different mediums should be considered.
