摘  要:目的建立从人胚胎干细胞向成熟嗜酸性粒细胞定向诱导分化的高效体外培养模型。方法采用人胚胎干细胞与小鼠主动脉-性腺-中肾(m AGM)基质细胞共培养的方法,进行早期造血干细胞诱导分化,将诱导分化的造血干细胞进行悬浮培养,在含有血清的培养基中,分阶段添加不同细胞因子组合进行成熟嗜酸性粒细胞的定向分化和增殖。结果获得从CD34+CD45+细胞扩增了约651倍,且纯度高达95%以上,CD88+Siglec-8+EPO+的成熟嗜酸性粒细胞。结论本研究建立了1种人胚胎干细胞高效定向分化为成熟嗜酸性粒细胞的模型,为进一步深入研究嗜酸性粒细胞的早期发育调控和相关疾病的发生机制奠定了基础。Objective To establish a model for inducing directional differentiation of human embryonic stem cells into mature eosinophils in vitro. Methods Human embryonic stem cells and mAGM-S3 (a routine AGM-derived stromal cell line) were first co-cultured to induce the development into early hematopoietic stem cells, then were induced into mature eosinophils in suspension culture with different cytokine combinations which were specific for proliferation and maturation "of human eesinophils. Results CD34 + CD45 + cells were induced to differentiate 651 times into CD88 + Siglec-8 + EPO + mature eesinophils with 95% purity. Conclusion This study established an efficient model in which human embryonic stem cells undergo directional differentiation into mature eosinophils which helps lay a foundation for further studies on mechanisms that control early developmental regulation of human eosinophils and related diseases.
