摘  要:目的评估钴60辐照对MAP保存液中红细胞体内清除相关指标造成的影响,寻找辐照促进红细胞在体内清除的可能机制。方法将滤除白细胞的红细胞悬液分3组:对照组不予辐照,即刻辐照组在采集后立刻辐照,14 d辐照组在保存14 d后进行辐照。在储存期的d1、7、14、21、28、35检测3组红细胞平均细胞体积(MCV)、红细胞刚性指数及丝氨酸磷脂(PS)。结果辐照后短时间内3组间MCV无差异,d14日起即刻辐照组与其它2组出现差异,且差异随保存时间的延长而增大。14 d辐照组在辐照前MCV升高缓慢,辐照后升高加速,但至d35时仍低于即刻辐照组。这种变化规律同样可以见于红细胞刚性指数。PS呈缓慢升高,存储至d28天3组才开始出现明显差异,即刻辐照组PS阳性率最高,对照组最低。结论钴60辐照可能通过影响平均细胞体积及红细胞变形性,促进红细胞体内清除,降低红细胞活力。辐照可促进丝氨酸磷脂缓慢外化,但在血液存储有效期内应该不是引起红细胞清除率增高的主要原因。Objective To investigate the effect of Co60 irradiation on the in vitro properties of red blood cells (RBCs) in MAP preservative solution, and to explore the mechanism through which irradiation promotes RBC elimination in vivo. Methods The leukoreduced RBC suspensions were divided into three groups. The control group was not irradiated; the immediate irradiation group was irradiated immediately after collection and the 14th day irradiation group was irradiated after 14 days' preservation. On the 1st, 7th, 14th, 21st, 28th, 35th day of storage, mean cell volume (MCV), RBC rigidity index and phosphatidyl serine (PS) of the three groups were tested. Results There was no difference in MCV between three groups shortly after the irradiation. From the 14th day on, the immediate irradiation group started to look different from the other two groups. On the 14th day, the MCV of irradiation group increased slowly before irradiation while rising sped up after irradiation, but the MCV was still lower than that of the immediate irradiation group until the 35th day. Same variation was seen in the test of RBC rigidity index. PS increased slowly and there were no significant differences among the three groups until the 28th day of storage. Conclusion Co60 irradiation may promote the RBC elimination in vivo by influencing MCV and RBC deformability. Irradiation can promote the exposure of PS on the outer plasma membrane of RBC, but it may not be the major cause of increasing RBC clearance rate during the validity period of blood.
